Cell Immuno-Chip (Cim-Chip) is a kind of Bio-Chip which could be used to detect the cell surface antigens by directly inspecting the cell with a microscope on the basis of protein chip and the specific reaction between the cell surface antigens the associated antibodies. Cim-Chip has the advantage of high throughput and could be used to detect up to several hundreds of cell surface antigens simultaneously. In this paper, the experimental conditions of Cim-Chip were optimized. The technique was used exploringly on the immnuophenotype of acute leukemia and the detection of cell tumour marker of the enteron cancer and won the approving result. To manage the vast data and images of the Cim-Chip, a set of software was programmed and named the detecting system of Cim-Chip. The major contributions include the following aspects:1.The fabricating conditions of the Cim-Chip were evaluated. CD45 monoclonal antibody was immobilized on a chip. After the reaction between the immobilized antibody and the leukocyte solution, the capture efficiencies of leukocytes were compared among the different cell concentrations, the different antibody fixing times and the different incubating times. The optimized experimental conditions established the experimental fundament for the investigation and appliance of the Cim-Chip.2.The exploration of using the Cim-Chip on the immunophenotye of acute leukemia. The associated antibodies of the M2a leukemia such as CD13,CD14,CD15,CD33,CD34,CD45 and HLA-DR were fixed on a chip. After the reaction between the immobilized antibodies and the leukocyte solution of M2a patient, the images were captured and compared with the results of the cytometry. A convincing coherence result between both methods was achieved.3.The exploration of the Cim-Chip on the detection of cell tumour marker of the enteron cancer. The associated antibodies of the enteron cancer such as P21, P27, P53, CD44V6-e, C-met, CDK4, HMM-7 and MG7 were fixed on a chip. The results demonstrated the possibilities of using the chip in the detection of cell tumour marker of the enteron cancer because all of the aboveantibodies had the positive binding point.4.The detecting system of the Cim-Chip was programmed. In order to manage the vast data of the Cim-Chip, a set of software was programmed and named the detecting system of Cim-Chip. The system could save all of the microscopic images and the associated information into the database. The system realizes the synchronous management of the images and the information.
