Expression Of OVCA1 Gene And Its Relation With The Expression Of CyclinD1 In Epithelial Ovarian Tissue

Objective To investigate the expression of OVCA1 gene in epithelial ovarian cancer and its relation with the expression of cyclinD1.Methods Expression of OVCA1 gene was evaluated by reverse transcription polymerase chain reaction ( RT-PCR) in 31 cases of epithelial ovarian cancer , and compared with that in 10 cases of ovarian borderline cancer ,10 cases of ovarian benign tumors and 10 cases of normal ovary tissue. Expression of cyclinD1 gene and cyclinD1 protein was detected by RT-PCR and immunohistochemistry S-P method, and the correlation between them was analyzed.Results1. Positive expression and relative content of OVCA1 mRNA was detected less in ovarian carcinoma(16.13%, 2.59±6.26)than ovarian borderline tumors(60%, 26.11±23.08) ,ovarian benign tumors (90%, 79.74±29.27)and normal ovary tissues(100%, 86.49±28.52).And there are significant differences between them(P<0.05).2. The expression of OVCA1 mRNA and its relative content in advanced clinical stage(Ⅲ,Ⅳ)were found significant differences to initial clinical stage(Ⅰ,Ⅱ) in ovarian carcinoma (P<0.05). And they were statistically less in those with lymphatic metastasis and ascites. In different histological grades, ages and histology classifications , there were no statistical difference.3. Expression of cyclinD1mRNA was growing while the expression of OVCA1mRNA was decreasing in ovarian carcinoma, borderline tumors, benign tumors and normal ovary tissues. There was negative correlation between them respectively (P<0.05). Positive expression of OVCA1mRNAwas 0% in ovarian carcinomas whose cyclinD1mRNA's expression was positive. While the expression was 41.67% in those whose cyclinD1mRNA's expression was negative. And the correlation between them was negative (P<0.05).Conclusion1. Expression of OVCA1 gene was less in ovarian cancer .It may reflects the progression of disease.2. Expression of OVCA1 gene was correlated with clinical stage, lymphatic metastasis and ascites. But there were no difference in histological grades, ages and histology classifications. It may reflect the invasion and metastasis.3. There was negative correlation between the expression of OVCA1 gene and cyclinD1 gene/protein.
