Pulmonary thromboembolism ( PTE) is from the venous system or right heart obstructive pulmonary thrombosis or its branch due to the disease, in the pulmonary circulation and respiratory dysfunction its main clinical and pathophysiologic features. Pulmonary embolism pulmonary embolism for the most common type, accounting for the vast majority of pulmonary embolism. Because of its pathogenesis of more hiding, no specific symptoms, clinical detection rate is relatively low, more cases of misdiagnosis, is the most important prognostic factor. In recent years the number of cases reported there is a growing trend, has gradually become a common lung disease. At present, the treatment of the disease mainly taken thrombolysis and anticoagulation, and the effects of pulmonary thrombolytic efficacy of the factors become critical. Based anticoagulant factor -- Tissue Factor Pathway Inhibitor (TFPI) start, research PTE lung tissue of TFPI and the variation of tissue factor (TF) relations.Objective : To establish a rat model of PTE, observed at different stages of deep vein thrombosis produced by the PTE and TFPI changes and TFPI and TF relations.Method :1. 72 Sprague-Dawley rats, half male and half female. stratified according to the male and female birds were divided into the experimental group (36) and the control group (36). Test groups, deep vein thrombosis time (1 day, 7 days, referred D) different thrombosis and pulmonary embolism after hours (1 day, 7 days, 14 days, or P) divided into D1P1. D1P7, D1P14, D7P1, D7P7, D7P14 six groups the control group on thecorresponding time is divided into six groups, a total of 12 teams, each comprising six.2. The experimental rats right femoral vein surgery, it formed deep vein thrombosis, Under different times removed femoral vein thrombosis injected into the contralateral femoral vein PTE model, based on the rats were sacrificed at different time, Retrieve lung tissue for testing.3. HE lung tissue was observed in lung tissue of rats.4. Successfully using immunohistochemical staining to detect TFPI, TF expression.5. The results assessment: Use cold CCD professional DP70 digital image analysis system to sections observation, membrane to the cytoplasm or brown granules emerged as a positive expression of each slice randomly selected five outlook use image pro plus 5.0 pathological image analysis system in the average gray value, take them as the arithmetic average of each slice of the average gray value, unit area TFPI, TF content.6. Statistically : Data are mean±standard deviation (mean±SD), randomly sampled 20 strains using statistical analysis software, the data were used linear correlation between the test and t test. Correlation coefficient r, 1≤r≤1 said two variables linear relationship, r positive number indicates there is a positive correlation between, r negative expressed negative correlation between P indicated statistical significance.Results :1. In vivo thrombus formation produced animal model pathologically proved to be a success.2. The experimental group D1P1 D7P1TFPI with TF and the relative value compared with the control group increased significantly, and the remaining group and the control group were not significantly different.3. Based on the experimental group TFPI and TF expression analysis found TFPI and TF positive linear correlation.Conclusion :1. Frozen sections by immunohistochemistry of semi-quantitative determination of TFPI, TF in the lung tissue of expression.2. Measured TFPI, TF in the lung tissue of found embolization in the first days of an experimental group than the control group was significantlyincreased, indicating the body early embolization coagulation and anticoagulation system has been activated.3. Based on the experimental group TFPI and TF expression analysis found TFPI and TF positive linear correlation.
