Objective: Utilize antitumor drugs to rat ileum, to observe antitumor drug inhibited mucus secretion of ileum beaker cells.Method: SD rats(weight 200—250g) were divided into three groups : mitomycin group, Idarubicin group, NS control group. Every rat was given dissociative ileum ansa interstinalis in vivo; thedrug's metrology is accord- ing to preliminary test. Then got the mucus of ileum ansa interstinalis at 8h,16h,24h,32h,40h,48h, and determine the content of mucus protein. Sacrificed rats after 48h, got ansa interstinalis and the five centimeter nomal ileum of superior extremity of andanastomotic stoma and stained with HE,AB-PAS,immunohistochemistry after paraffin imbedding, to observe the mucus secretion of beaker cell and the change of beaker cell.Result:1.Effect of histomorphology: There is no obviously injuryof mucosa and submucosa, experiment group has no obviously difference compared with control group.2.Effect of secretion of beaker cells Detected the protein level of each time stage using BCA method. Mucus level of mitomycin group is less than NS control group. Mitomycin group's mucus level is less than NS control group at 32—40h ,40-48h.There was no significant difference between Idarubicin group and NS control group.3.Effect of amount of ileum beaker cells The number of beaker cells has obviously difference between mitomycin group and NS control group, but Idarubicin group has no difference with control group.4. Immunohistochemistry of mucoprotein MUC2 The number of beaker cells that have buffy grana was counted from 5 fields of vision inevery sample. We got the same result from immunohistochemistry compared with AB-PAS. The number of beaker cells has obviously difference between mitomycin group and NS control group, but Idarubicin group has no difference compared with control group. 5. Effect of amount of the normal enteromucous beaker cells The number of beaker cells has no difference between NS control group and nomal group, but mitomycin group has obviously difference with normal group.Conclusion:1. Low dose chemotherapeutics does no damage to ileum ansa interstinalis'mucousa and submucosa2. Mitomycin inhibite beaker cells secreting mucus in dissociative ileum ansa interstinalis in vivo.3. Mitomycin had been irrited only effect of the dissociative ileum ansa interstinalis in vivo but don't of the normal intestinal tract.
