Objective: To study the development of mobilized autologous peripheral blood mononuclear cells in the enviroment of liver fibrosis and observe the effect of autologous peripheral blood mononuclear cell transplantation on liver fibrosis.Methods: Rat liver fibrosis was produced by injecting carbon tetrachloride subcutaneously. Rats with liver fibrosis were divided into four groups: recombinant human granulocyte-colony stimulating factor(rhG-CSF) mobilization group, atutologous peripheral blood mononuclear cell transplantation group(APBMCT), spontaneous recovery group and control group. rhG-CSF mobilization group was only administered with rhG-CSF . APBMCT group went through rhG-CSF mobilization and autologous peripheral blood mononuclear cell transplantation. Spontaneous recovery group underwent no management. The fibrosis rats sacrificed instantly after the last time of carbon tetrachloride administration were as the control group, sera and livers of control group were collected for latter measurement. After 6 weeks, the rats of all the other three groups were sacrificed and their sera and livers were collected. The ALT,AST,ALB,TBIL of all the four groups were measured and the livers of all the four groups were sectioned for pathological alnalysis. In APBMCT group, immunofluorescent method was employed for the observation of the development of autologous peripheral blood mononuclear cells in the enviroment of liver fibrosis.Results: Compared with control group, the ALT,AST,ALB,TBIL and the representaion of liver pathology of rhG-CSF mobilization group, APBMT group, spontaneous recovery group were improved(p0.05). Compared with spontaneous recovery group, the ALB,TBIL and the representation of liverpathology of rhG-CSF mobilization group, APBMT group were improved(p<0.05). There was no significant difference between rhG-CSF mobilization group and APBMT group both in biochemistry and liver pathology. Autologous peripheral blood mononuclear cells labeled with PKH26-GL expressed albumin in fibrosis liver of rat.Conclusoins: Rat liver fibrosis induced by injecting carbon tetrachloride subcutaneously has the tendency of spontaneous recovery after discontinuation of injection. rhG-CSF can accelerate the recovery of rat liver with liver fibrosis induced by injecting carbon tetrachloride. Mobilized autologous peripheral blood mononuclear cells can differentiate into cells which can express albumin.
