In order to develop and utilize Phellinus Lineus resources, the main biologic effects of polysaccharide of Phellinus Lineus were studied on three aspects: anti-tumor proliferation, anti-tumor migration and anti-angiogenesis. The basis data on studying the active composition of polysaccharide and the mechanism of its anti-tumor effects suggested that Phellinus Lineus is an effective medicine for curing tumor.To identify the anti-tumor proliferation effect in vivo, nude mice modle was established for this study. It is observed that the tumor volumn is far smaller in the PL treated nude mice than in the PBS treated ones. The inhibition rate is 58.94% in HO-8910PM injected mice, and 73.36% in Bcap-37 injected mice. Meanwhile we found the apoptotic tumor cell on the frozen-slice. These data illuminated that PL is an effective drug for control the tumor growth both in vivo.By MTT assay in vitro, the ability of anti-tumor proliferation by polysaccharide extracted from Phellinus Linteus were studied by HO-8910PM cell line and Bcap-37 cell line. The effect is significant by treating the cell with polysaccharide extract for 48h. Then three kind of cell line, HO-8910PM, Bcap-37 and HT-29 were treated with PL for 48h. For indicating the mechanism of anti-tumor growth effects by PL, some flow cytometer (FCM) research had done. It is detected that the cell cycle was blocked in G0/G1 phase with the PL treatment for 48h, and while prolonging the PL treatment time to 96h, a apoptotic peak was observed. Thus it can be seen that arosing the change of cell cycle is one of the mechanisms for anti-tumor, and prolonged PL treating can lead the cell to apoptosis.The anti-tumor migration effects by PL were studied in vitro. The study data showed that PL can reduce the adhesion of tumor cell to both extracellular matrix (ECM) and HUVEC, the adhesion rate can be reduced by 17.68% with the exposion of 250μg/ml PL. We took a drawing line assay and a Transwell assay to evaluate the PL effects on tumor cell migration. Both methods proved than PL can inhibite the migration ability of tumor cell. For tumor cell invasion research, transwell was once more used with Matrigel, and it is found 167μg/ml and 500μg/ml PL could inhibite the cell invasion by 19.11% and 36.99%.The anti- angiogenesis effect was studied in vitro. Since the procedure of tumorproliferation and migration goes inevitably with the formation of new vein, scientists suggests that it is also an efficient approach to anti-tumor by controling the formation of new vein. The results turned to be that PL can indeed inhibite both the proliferation of HUVEC and its vessel forming. And the CAM assay also indicated the anti-angiogenesis effect of PL.
