| Objective: To explore the mRNA expression of p53 splice variants:Δ133p53, Δ 133p53 β , Δ133p53 γ , p53 β and p53 γ in renal cell carcinoma tissues and their corresponding adjacent non-cancerous tissues and investigate their roles in carcinogenesis.Methods: Fifteen pairs of fresh renal carcinoma tissues and their corresponding adjacent non-cancerous tissues were obtained from radical nephrectomy in the Seconed Hopsital of Shandong University between May 2004 and December 2006. All the samples were examined for mRNA expression levels of p53 splice variants:Δ 133p53, Δ 133p53, Δ 133p53 γ, p53 β and p53 γ by reverse transcription polymerase chain reaction (RT-PCR).Result:1. Δ133p53 was expressed in 13 out of 15 non-cancerous tissues (13/15) and in 10 out of renal carcinoma tissues (10/15), but there were not significant differences between 15 pairs of fresh renal carcinoma tissues and their corresponding adjacent non-cancerous tissues (P>0.05).2. Δ133p53 3 was not expressed in both renal non-cancerous tissues and carcinoma tissues.3. Δ 133p53 γ was detected in 4 out of 15 renal non-cancerous tissues (4/15) and in 1 out of carcinoma tissues (1/15), but with no significant differences between carcinoma and non-cancerous tissues.4. p53 3 was detected in 3 out of 15 renal non-cancerous tissues(3/15) and in 10 out of carcinoma tissues(10/15) with significant differences between carcinoma and non-cancerous tissues (P<0.05). The expression level in fresh renal carcinoma tissues was much higher than their corresponding adjacent non-cancerous tissues.5. p53 γ was 'detected in 12 out of 15 renal non-cancerous tissues(12/15) and in 4 out of carcinoma tissues(4/15) with significant differences between carcinoma and non-cancerous tissues(P<0.05). The expression level in fresh renal carcinoma tissues is lower than their corresponding adjacent non-cancerous tissues. Conclusions:1. Δ133p53, Δ133p53 γ, p53 β , p53 γ except Δ133p53 β can be detected in both renal non-cancerous tissue and carcinoma tissue.2. In both renal non-cancerous tissue and carcinoma tissue, expression of p53 β and p53 γ have significant difference.3. The differential expression of the p53 isoforms in human renal carcinoma may be related to the growth, proliferation and metastasis of renal cell carcinoma. |
