Objective To evaluate the feasibility of diode laser-inducing choroidal neovascularization (CNV) in Brown Norway (BN) rats with detect of fundus fluorescein angiogrphy(FFA) and optical coherence tomography (OCT)and if octerotide and octerotide combined to transpupillary thermotherapy (TTT) endostatin might inbibit the development of experimental CNV of BN rats.Methods The eyes of experimental rats were received by diode laser diode laser. chloroacetophenone (100mg/kg) were injected in the venter of the choosed 10 BN rats, Ten diode laser spots (150mW, 75 μm, 0.1s) were applied to one fundus around the optic disc of rats. On 7, 14, 21 and 28d after photocoagulation, 5 BN rats were selected randomly, The same spot of CNV formation was determined by FFA and OCT after 20% sodium fluorescein (0.5ml/kg) were injected in the venter of rats. the change of number/area of fluorescein leakage (mm~2) of the early-phase (<2 minutes) and late-phase (>10 minutes) of each day were compared.20 BN rats were grouped to 4 groups after photocoagulation: 5 was considerd as blank control,the other 15 BN rats as the treatment group, group 1 were received medication on the day of photocoagulation, group 2 received on 14d after photocoagulation , group 3 received medication and TTT on 14d after photocoagulation .drug: octreotide, 50μg/kg, retrobulbar injection, q7d*2;TTT: spots:0.4~0.54mm, power :56~76mW, time: 60s, once. on 7, 14, 21 and 28d after laser photocoagulation, 20% sodium fluorescein (0.5ml/kg) were injected into the venter of rats. The same spot of CNV formation was determined by FFA and OCT and the intensity of fluorescein leakage were graded, The experimental data were analyzed with ANOVA and LSD. P<0.05 is thought to have statistically significant difference.Results1. Study of experimental CNV model1. 1 the lesions showed irregular fluorescein leakage on 7d after photocoagulation, and the number increased on 14d,to peak on 21d , the incidence of CNV was 36%, 55%, 69% and 70% respectively on 7,14, 21 and 28d.The number /mean area of fluorescein leakage had no statistically significant difference between the early FFA phase of 14,21 and 28d (P>0.05), but significant differences were found when 14,21 and 28d compared to 7d (P<0.05). The number of fluorescein leakage of early-phase of 7,14,21 and 28d was not significantly different from late-phase (P>0.05).1. 2 The correlation of the mean area of fluorescein leakage and the mean retinal thickness was obvious (r=0.73, P<0.05)1. 3 Complication:2/10 eye 2/20 lesion showed subretinal hemorrhages.2. Study of octreotide and octreotide combined to TTT on the experimental CNV.2. 1 Group 1 was found no fluorescein leakage on 7 and 14d, OCT show focal neurosensory retina were fragmented and shallow detached. There were distinctly statistical differences of grade of FFA and retinal thickness between group 1 and control (P<0.05).2. 2 Group 2 showed fluorescein leakage but weaker than control, and weak fluorescein leakage spots amount to 35% (18/44) .there were no statistical differences compared to control on 21 and 28d (P>0.05).2. 3 Group 3 showed little fluorescein leakage and weak leakage spots accounted to 35% (18/44) , OCT showed CNV became thin , even disappeared. Group 3 also showed a significant reduction in the grade of FFA and retinal thickness compared with control (P<0.05), but had no statistical differences compared to Group 3 (P>0.05).2. 4 The overt adverse effects and complications were not found in the treatment of choroidal neovascularization with octerotide and octerotide combined to TTT. (P<0.05).Conclusion:1. Diode laser photocoagulation could be successfully used to produce CNV experimental model in the BN rats. The check of both FFA and OCT show CNV completely formed on 14d after photocoagulation .2. The belly injection of sodium fluorescein was easy and the image of FFA was legible.3. FFA combined to OCT was a objective and effective method to detect the formation and change of CNV.4. Intrabulbar injection of octerotide could prevent the formation and development of experimental CNV , it was essential to use in early stage.5. Octerotide combined to TTT could availably suppress the development of experimental CNV.6. The way of octerotide and octerotide combined to TTT in the treatment of experimental choroidal neovascularization were safe and valid.
