The Study On Manganese Chloride Toxicity Effect To PC12 Cells And Its Mechanism

ObjectiveTo study the mole-mechanism in the apoptosis of PC12 cells induced by manganese chloride by using PC12 cells as the model of dopaminergic neuron, examing the cells growth, livingness and apoptosis of PC12 cells in various doses of manganese chloride and studying the significance of the expression of P53, MDM2 proteins in the apoptosis of PC12 cells. And the contradict effects of astragalus, providing further data for manganese chloride neurotoxicity and protective ways about it.MethodsUsing PC12 cells as the model of dopaminergic neuron; MTT colorimetry test were performed to exam cell growth and livingness of PC12 cells in various doses of manganese chloride; Manganese chloride induced apoptosis and the contradict effects of astragalus in PC12 cells were observed by flow cytometry (FCM) and TUNEL; And immunocytochemistry was used to measured the expression of P53, MDM2 proteins.Results MTT results found that manganese chloride might suppress the proliferation of PC12 cells in dose and time-dependent manners and induce PC12 cells apoptosis , it has dose-dependent manners; and it has some contradict effects of astragalus in the apoptosis by manganese chloride in PC12 cells; immunocytochemistry showed that manganese chloride boosted the expression of P53 proteins and inhibited the expressions of MDM2 proteins and also have dose-dependent manners. There are positive correlation and negative correlation between the expression of P53, MDM2 proteins and manganese induced apoptosis in PC12 cells.Conclusions(1) Manganese chloride can inhibited the growth and livingness of PC12 cells in a certainty condition;(2) PC12 cells were induced to apoptosis when they exposed to Manganese chloride by boosting the expression of P53 proteins and restraining the expression of MDM2 proteins and have dose-dependent manners;(3)There are some contradict effects of astragalus in the apoptosis by manganese chloride in some extent dose in PC12 cells;...
