Objective: Many investigations had confirmed that both amiodarone and magnesium sulfate were broad-spectrum antiarrhythmic drugs. Amiodarone is Class III antiarrhythmic drugs, moreover, it had been confirmed the best effective and safe possibly. In this study, the patients with refractory premature ventricualr contraction were treated by oral amiodarone combined with magnesium sulfate injection in order to observe the clinical effects and safety.Methods: 45 patients were randomly divided into two groups, control group (oral amiodarone longly) and curative group(oral amiodarone combined with magnesium sulfate injection)in which 25% magnesium sulfate 2.0g was dissolved in 250ml of 5% glucose or 0.9% natrium chloride, once a day for one week. The methods of taking oral amiodarone as follow: 0.2g, three times a day in the first week, 0.2g, twice a day in the second week, once a day in the third week as well as afterward.Results: The decrease of the number of premature ventricualr contraction and the times of temporal ventricular tachycardia and theimprovement of heart function, all of above had significant differences compared post-treatment with pre-treatment, p<0.05. Curative group was better than control group, p<0.05. Compared post-treatment with pre-treatment, there was also significant difference in heart rate slowed, p<0.05, but no difference between two groups. There were no changes in the blood pressure and biochemistry index. No side-effects had happened, such as torsade de pointes or pulmonary interstitial fibrosis or dysthyroidea or neurotoxicity etc.Conclusion: Oral amiodarone combined with magnesium sulfate injection was better than oral amiodarone longly not only in the decreased of refractory premature ventricualr contraction but also in the improvement of heart function, which indicating their synergetic effects. The application was safe in clinic.
