Objective: Progestin receptor antagonist was suggested enhancing estrogen and smooth muscular contraction of oviduct. Hypothesis is that mifepristone, a progestin receptor antagonist, increases oviduct contraction, reduces tubal inflammatory process, further, and treats in clinical practice barrenness caused by tubal pathologic conglutination. The present study was to prove the hypothesis that mifepristone enhanced contractile frequency of tubal smooth muscles in vitro recorded by bio-physiologic experimental system. Morphological alteration after mifepristone treatment in animal models which were implanted with staphylococcus, a clinically potential pathogeny, and suffered acute tubal inflammation was performed with macroscopic and microscopic observation and analysis. Mating and pregnant ratio were undergone in some animal models followed mifepristone treatment. The aim of this study is to find new methods to treat barrenness caused by tubal pathologic conglutination.Methods: Oviducts (n=5) were taken from paunch rabbits and separated into 2 cm long, and then hanged on 3.5 ml tissue glass bath (HW-400S, TM, Chengdu, China) filled with Kerbs solution (in mM: 118 NaCl, 2.5 CaCl2, 1.2 MgSO4, 24.9 NaHCO3, 1.2 NaH2PO4, 4.7 KC1, and 12.2 glucose) that was maintained at 37.8℃ and aerated continuously with 95% O2-5% CO2. Isometric contractile force generated by oviduct by a force-displacement transducer (JH-2, Institute Space Medical-Engineering, Beijing, China) and recorded the response continuously on a polygraph (BL-420E, TM, Chengdu, China). After basic physiologic recording, mifepristone was administrated in Krebs solution until 10μM, and subsequent effect on tubal smooth muscle recorded.
