Objective: To investigate the mechanisms of anoxia/reoxygenation and the effects of Fosinoprilat on the culture cardiocytesMethods: Cultured cardiocytes of neonatal SD rats (1-3d)were randomly divided into five groups: control group; anoxia reoxygenation (A/R) group; Fosinoprilat1(F1+A/R)group; Fosinoprilat2(F2+ A/R)group and Fosinoprilat3(F3+A/R)group. In each group, the cardiocytes were subjected to anoxia (3hr) followed by reoxygenation (1hr) on the model of the cultured myocardial cells. At the end of experiment, cardiocytes beating rate, cell viability and the activity of LDH in culture were measured;furthermore, The intracellular dissociative calcium concentration of cardiocytes was measured by Flow Cytometer (FCM) and the expression of SERCA2 mRNA was measured by RT-PCR.Results: (1). Beating rate of cardiocytes: The beating rate was significantly slow in A/R group(11.50±3.39beat/min) compared with that of control group (99.83±4.22 beat/min) (p<0.01) ; Between each group of Fosinoprilat and control group, the beating rate was similar to each other (p>0.05) ; Compared with A/R , the beating rate of each group of Fosinoprilat was significantly increased (p<0.01) .
