Objective To verify the cccDNA in chronic hepatitis B patients' sera is an signal Of the liver damage; To study the clinical and pathological relationship between the quantitation of HBV cccDNA and chronic hepatitis B(CHB).Mthods selected Randomly 20 slight and mild patients with chronic hepatitis B,20 patients with severe chronic hepatitis and 20 HBV viral carriers(all patients were HBVDNA positive).The cccDNA of the 60 invesgated patients were detected by selective real-time fluorescent quantitative PCR, HBV DNA were detected by routine PCR, and the ALT , AST and Tbil were detected by routine method. The 10 patients among those were punctured to obtain liver biopsies. liver biopsies were used for pathology inspection and the cccDNA, HBVDNA detection by real-time fluorescent quantitative PCR and routine PCR. Results There was significantly different in the quantify of three groups'cccDNA in sera(p=0.000 < 0.01). The serum cccDNA level of chronic hepatitis B patients was correlated with the level of ALT ( Rs=0.612 p=0.000 < 0.01 ), with AST ( Rs=0.632 p=0.000 < 0.01), with Tbil(Rs=0.337, p=0.008 < 0.01,but Rs<0.4). In the 10 patients with liver puncture,the value of cccDNA in liver biopsies and sera : 4.33±1.27,4.67±1.30;HBVDNA in liver biopsies and sera: 2.61±0.88, 3.72±0.88. The cvalue of ccDNA in liver biopsies was correlated with the value of HBVDNA in them and sera and the value of cccDNA in sera(R=0.766,P=0.01<0.05; R=0.731,P=0.016<0.05;...
