The incidence rate of head and neck cancer is approximately 14/100,000, occupied the whole body malignant tumor 16%-40%, and the 5 years survival rate is 35%-60% on the whole. The dissection of head and neck is complicated and the organ is crowded, so these cause the surgery to cure the diseases difficulty ; the head and neck cancer is not sensitive to the radiotherapy and the chemotherapy , so some patients lose the opportunity of healing well. Therefore the gene therapy is a very good substitution method to cure the head and neck cancer.The gene therapy is to transfer the human normal gene or the functional gene to the target cell by a certain way in order to correct the defective gene or to pruduce the treatment function, thus achieved a new biomedicine technology that can cure disease. The gene therapy must solve three key questions to obtain the breakthrough: the highly effective gene delivery system, the controllability of gene expression and the better functional gene.The gene delivery system is the core technology of the gene therapy, and it may be divided into two parts: the viral vector system and the non- viral vector system. In the recent 20 years, only the minority several kinds of viruses like retrovirus (including HIV), adenovirus, adeno-associated virus, herper virus((including herpes simplex virus, poxvirus officinalis and EB virus) and alphavirus are successfully modified as the gene delivery vector and have developed the varying degree application.Adenoviruses are attractive vectors for cancer due to their unparalleled capacity for gene transfer, stability in vivo, and the production of high titers of current good-manufacturing-practice quality adenoviruses is established.
