It is a top topic of tumor vaccine research to study tumor vaccine based on MUC1 target at present. MUC1 is the mucin that is a membrane protein composed of core peptide and carbohydrate side chain expressing on normal epithelia cells and adenocarcinoma cells, and its extracellular domain consists of tandem repeat sequences of 20 amino acids (SAPDTRPAPGSTAPPAHGVT). There are some differences of MUC1 expression between normal and tumor cells. Normal MUC1 exists on ductal epithelia as a large heavily glycosylated protein is expressed only on the apical cell surface directed the duct lumen. The expression of MUC1 is up-regulated on tumour cells where it undergoes changes in deglycosylation and distributes on whole tumor cell surface, that make it distinct from normal mucin. Therefore, the tumor-specific epitopes hidden in the normal of MUC1 are exposed, that make it a novel target for active tumor immunotherapy. It has been showed that the sequences of PDTRP on MUC1 core peptides can be recognized by kinds of monoclonal antibody against MUC1. Moreover, it is more important that the sequences can be recognized by cytotixic T cells (CTL) and lead to kill tumour cells by restrictive or non-restrictive MHC.
