Objective: To design and develop a nisoldipine buccal adhesive tablet to overcome the drawbacks Of the conventional dosage forms such as the high first-pass metabolism, fluctuated blood level, frequent dosing and the lower bioavailability. We also studied on the Pharmacokinetics,relative bioavailability and in vitro-in vivo correlation of the nisoldipine buccal adhesive tablets.Method: nisoldipine buccal adhesive tablets were prepared by the direct compression method using carbopol( CP )974 and chitosan (CS)as bioadhesive matrix in combination with lactose as pore formater and sodium deoxycholate as permeation enhancer.The buccal adhesive tablet was designed into the double-layered structure comprising the protective layer and the drug-containing one to provide drug delivery in an unidirectional fashion to the mucosa and to avoid loss of drug due to wash-out by saliva .Based on the single tests , four important factors were chosen that affected both the drug release rate and the...
