Objective: To evaluate the empirical method of myocardial transplantation. Moreover ,we want to study the effect of rhaFGF and autologous skeletal muscle satellite cells (MSCs) on transplantation to acute myocardial infarction(AMI) adult rabbits.Methods: This study included three parts.First, autologous MSCs were divided into small tissue fragments and cultured in vitro. Second, AMI model was established with bipolar electrocoagulator through blocking the left anterior descending coronary artery(LAD).Third,our 64 adult rabbits were randomly allocated to four groups: Group A (control group), Group B(MSCs transplanted group), Group C(transplanted with rhaFGF and MSCs), and Group D(rhaFGF injected group). RhaFGF and the autologously MSCs were injected into the infarcted area. To evaluate curative effect of different methods, we chose continuous electrocardiographic(ECG)monitoring, pathologic section study, echocardiographic (UCG) study and infarcted area analysis to act as indicators.Results: (1) The cells obtained from culturing tissue fragments showed to have strong proliferative ability in the culture solution. We had checked the purity of MSCs was over 90%, through α-actin staining. (2) Successfulness of establishing AMI model with bipolar electrocogulate was confirmed by continuous ECG monitoring and pathologic section analysis. (3)Two months after transplantation, the infarcted area measurement showed there were significant difference in group C vs group A(P<0.01), and group D vs group A(P<0.01). There was no significant difference in group B vs group A(P>0.05). (4)Two months after transplantation, ejection fraction(EF) of UCG study showed there was significant difference in groupB, groupC, and groupD vs groupA respectively(P<0.01). (5)Through immunohistochemistry analysis, we found that connexin43 staining of the cells in infarcted areas of group A showed negative, while those from group B, group C and group D showed weakly positive to positive.
