Objective To evaluate the efficacy and safety of zoledronic acid injection for the pain relief in patients with metastatic bone cancer.Methods A total of 56 patients were divided into group A(study group) and group B(confrol group), 28 patients in each group, and received zoledronic acid and pamidronate disodium for treatment respectively. 28 patients received intravenous infusion of zoledronic acid (4mg, treatment group) for 15 minutes; and 28 patients received pamidronate (90mg, control group) for 4 hours. The patients were followed up for 28 days to assess the efficacy and safety of the single dose treatment.Results The response rate was 71.4% in the group A, with 21.7%(6/28) CR and 52. 2 %(12/23) PR While the response rate was 64.3% in the group B, with 13.0%(5/28) CR and 47.8%(11/23) PR. There was no significant difference between the two groups (P> 0.05). In group A and group B, the mean time to pain relief was (3.96±2.26) days and (4.43±1.79) days respectively, and the mean duration of the pain relief was (27.68±1.89) days and(14.60±2.94) days respectively , with no significant difference between the two groups (P<0.05).No significant differences were found in the improvement in quality of life (78.6% versus 75.0%, respectively). The total incidence of adverse effects was 53.6% and 60.7% respectively, including fever, flu-like symptom, temporary muscle and skeletal pain and nausea. All of the adverse effects were mild and no intervention was involved. No severe side effect was found in both groups. There was no significant difference in the incidence of adverse effects in the two groups (P>0.05).Conclusion The effect of malignant osseous metastasis pain treated with Zoledronic acid injection was obvious. The treatment response could be persistent for a long time, and the drug is easy to be used in the clinic and has excellent safe profile.
