| Schizophrenia is a very complicated disease,it refers a lot of fields. now,its pathogeny is being studied,a great number of scholars consider that it is basically related to heredity,neural development,neurotransmitter,at the same time,other factors such as mentality,family,community etc also play significant roles.Although pathogenesis of schizophrenia has being studied,it is still Completely unclear.Heme oxygenase( HO)is rate-limiting enzyme degradating heme,the enzymilisising product of HO include biliverdin,CO and dissociative ferrum ( Fe2+) .Bilirubinic decomposition product has more powerful antioxidation than vitamin E;dissociative ferrum also plays roles on harmful product such as hydroxy radical and superoxide anion etc;As an important signal molecules,CO play significant roles on neuron information transfer and vascular tone regulation;people pay attention to HO,which is the main enzymatic system producting endogenous CO increasingly.Research in recent years has approved that endogenous gas signal molecules of H2S is similar with the two kinds of endogenous gas signal molecules of CO and NO,they are very important bio-regulating substances,and they have also participated in multiple kinds of physiological and pathological processes of central nervous system. SAM,activator of CBS which can catalyze H2S,has been proved to have antianxiety clinically.Therefore,we consider that H2S play significant roles during pathology of schizophrenia.Autoimmunity hypothesis of schizophrenia figures schizophrene has a certain degree of immunological functional disturbance,Hypothalmu- pituitary- adrenal(HPA) axis can produceimmunosuppression. Hydrocortisone,the production of HPA axis,serve as important medium of immunosuppression, so we presume that hydrocortisone possibly induce immunological dysfunction of schizophrene.In addition,HO-CO system and H2S correlate with hydrocortisone.HO-CO System can promote HPA to release CRH and ACTH,and then promote hydrocortisone releasing.In the occasion of stress, H2S can selectively inhibit CRH releasing,and then diminish hydrocortisone to keep the balance of neuroendocrine.To investigate relationship of HO,H2S,hydrocortisone and schizophrenia,we detect activity of HO and concentration of H2S,hydrocortisone in plasma,in order to provide experimental foundation for pathogenesy of schizophrenia.objective: Investigate level and clinic significance of HO,H2S an d hydrocortisone in schizophrenia prior and post risperidone treatment.methods: 28 schizophrenic clinical symptoms are scored through scale for the assessment of positive symptoms and negative symptoms prior and post six-week treatment.Activity of HO and concentration of H2S in plasma are detected by Spectrophotometer, concentration of hydrocortisone is detected by Electrochemiluminescence immunoassay(ECLI),compared with 28 control subjectives.Drug action on the activity of HO,the concentration of H2S and hydrocortisone for schizophrene is observed.results: Activity of HO of schizophrene in plasma prior-treatment is (997.51±79.75)nmol/h*l,it is lower than that post-treatmen(t1511.40±186.07)nmol/h*l and control subjectives(1633.27±259.03)nmol/h*l(P<0.05),Activity of HO post six-week treatment is uniform with control subjectives(P>0.05).Activity of HO of schizophrene in plasma has obvious negative correlation with score of scale for the assessment of positive symptoms and negative symptoms(r=-0.677 , P<0.05).Concentration of H2S of schizophrene in plasma prior-treatment is(87.22±5.12)nmol/L,it is more than that post-treatmen(t74.56±3.02)nmol/L and control subjectives(73.49±2.19)nmol/L(P<0.05),Concentration of H2S of schizophrene in plasma post-treatment is uniform with control subjectives(P>0.05),concentration of H2S of schizophrene in plasma has obvious positive correlation with score of scale for the assessment of positive symptoms and negative symptoms (r=0.965 ,P<0.05).Concentration of hydrocortisone of schizophrene in plasmaprior-treatment is(17.39±2.59)ug/dl,it is more than that post-treatment(12.42±5.70 ) ug/dl and control subjectives ( 12.80±6.62 )ug/dl(P<0.05),Concentration of hydrocortisone of schizophrene in plasma post-treatment is uniform with control subjectives(P>0.05),concentration of hydrocortisone of schizophrene in plasma has obvious positive correlation with score of scale for the assessment of positive symptoms and negative symptoms (r =0.589,P<0.05).conclusion:Low activity of HO of schizophrene in plasma and high concentration of H2S,hydrocortisone of schizophrene in plasma play significant roles on onset of schizophrenia.
