Objective: Kidney as a hypertransfusion organ, is extremely sensitive to ischemia. Ischemical reperfusion injury (IRI) is liability take place in the process of shock anabiosis, crush syndrome decompression and renal transplantation. Renal IRI is the important link of acute renal failure ( ARF), also the principal factor that influence the renal graft function recovery in morning and its long-term surviving in the renal transplantation. The pathology and physiology mechanism of renal IRI are multiplicity. Now, the most study is concentrated on active oxygen (ROS), heterophil granulocyle, complement system and so on, have deficient congnition of the meaning of aquaporins in this process. So this investigation raise the rat renal IRI model , study the changes of aquaporin-1,2 and the relationship between the changes of aquaporin-1,2 and the renal function in this process, hope probe the etiopathogenesis of the renal IRI and ARF in a new way.Method: Raise the rat left renal 40 minutes IRI model. Divided the experimental animal in control and ischemia reperfusion 1,2,3,5,7 day group randomly. Keep the urine of model rat to do urine detection and kill the rats on above-mentioned different time, keep the serum specimen to do renal function detection. Keep the renal collection to do HE coloretur,RT-PCR,immunohistochemistry and Western blot examination.Results: 1.The urine changes (1)The urinary production changes between the control groups did not have statistically significance(P>0.05). The ischemical reperfusion injury rats, urinary follow was significantly increased compared with the sham-operated rats, and reach the maxlmum at the third day after the operation (P<0.01).Then grow downwards gradually,became normal at the 7 th day after the operation(P>0.05). (2) The urine osmotic pressure changes between the control groups did not have statistically significance(P > 0.05 ) .The urine osmotic pressure of ischemical reperfusion injury group decrease obviously compared with the sham-operation group, and reach the maxim at the third day after operation (P<0.01), then increase gradually and become normal at the seventh day after operation(P>0.05).2.The changes of the renal function . The renal function changes between the control groups did not have statistically significance(P>0.05)The carnine, urea nitrogen of the IRI rats step up obviously compared with sham-operation group, and reach the maxlmum on the third day after the operation(P<0.01), degrade gradually from then on, and become normal on the seventh day after the operation(P>0.05).3.The kidney histology changes of IRI rat. The renal tubular epithelical cell of the IRI rats engorgement, necrosis and ablate manifest than control group.4.The mRNA expression level changes of AQP-1, AQP-2 The mRNA expression level of AQP-1, AQP-2 changes between the control groups did not have statistically significance(P > 0.05 ) The mRNA expression level of AQP-1, AQP-2 of IRI rats renal is manifest cut down compared to sham-operation rats. AQP-1 reach the lowest at the first day after IRI(P<0.01),then step up gradually ,and return to normal at the fifth day after the operation(P>0.05); AQP-2 reach the lowest at the third day after the operation(P<0.01) ,then step up gradually ,and return to normal at the seventh day after IRI(P>0.05).5.AQP-1 and AQP-2 proteinology examination The proteinology examination of AQP-1, AQP-2 changes between the control groups did not have statistically significance(P > 0.05 ) Immunohistochemistry and Western blot had used to detect the change of AQP-1, AQP-2 .They all cut down after the operation, AQP-1 reach the lowest at the first day after IRI(P<0.01), then step up gradually and return to normal at the fifth day after IRI(P>0.05); AQP-2 reach the lowest at the third day after IRI (P<0.01), then step up gradually and return to normal at the seventh day after IRI (P>0.05).Conclusion: 1.When the rats′renal had IRI happened, renal tubular epithelial cell necrosis, ablate obviously on pathology, 24 hour urinary production, carnine, urea nitrogen step up, urine osmotic pressure cut down. It is cues that IRI is one of the motivation of acute tubular necrosis and acute renal failure.2.The IRI rats kidney had the symptoms of hydrouria, urine osmotic pressure cut down and so on. Also had AQP-1, AQP-2 expression level cut down. It is cued that after IRI the AQP-1 and AQP-2 expression level decrease induce malabsorption of water, maybe one of a important factor of hydrouria, urine osmotic pressure cut down induced by acute renal failure.3.The IRI rats kidney developed acute renal insufficiency meanwhile had the AQP-1,AQP-2 changes followed , hinted that the changes of AQP-1,AQP-2 expression maybe the important indexes of renal function. They take all-important effect in the pathology and physiology process of renal ishemical reperfusion injury.
