Objective: To reveal the molecular mechanism of cancer metastasis by exploring the effect of N-cadherin and E-cadherin on the behavior of tumor cells with different mode of metastasis.Methods: A2780 and A549 cells were transfected with N-cadherin,E-cadherin and empty vector respectively( A2780N, A2780E, A27803.0, A549N, A549E, A5493.0), and screened by G418. Examine proliferation ability, adhesion ability, migration ability and survival ability of different cell lines with cell proliferation experiment, cell detachment experiment, cell adhesion rate experiment, cell-matrix adhesion experiment, clone formation experiment and scarification experiment.Results:1. Effects on A2780 of N-cadherin and E-cadherin: compared with the control group of A27803.0, the detachment rate and the clone formation rate of A2780N were both increased; the cell-matrix adhesion rate, the adhesion rate and the migration distance of A2780N cell line were all decreased. All the differences were statistically significant(P<0.05). The proliferation rate of A2780N was not significant different with A27803.0. Compared with the control group of A27803.0, the proliferation rate, the adhesion rate and the clone formation rate and of A2780E were all increased(P<0.05); the other indexes were all not significant different with A27803.0.2. Effects on A549 of N-cadherin and E-cadherin: compared with the control group of A5493.0, the proliferation rate and detachment rate of A549N were decreased; the cell-matrix adhesion rate, the adhesion rate, the clone formation rate and the migration distance of A549N were all increased. All the differences were statistically significan(tP<0.05). Compared with thecontrol group of A5493.0, the proliferation rate, the detachment rate, the adhesion rate and the clone formation rate of A549E were all decreased; cell-matrix adhesion rate of A549E was increased. All the differences were statistically significant(P<0.05). The migration distance of A549E was not significant different with A5493.0.Conclusion:1. The effects of N-cadherin and E-cadherin on the behaviors of ovarian cacer cells A2780 and lung adenocarcinoma cells A549 were different.2. N-cadherin could decrease the cell-matrix adhesion ability and cell-cell adhesion ability of A2780 cells; E-cadherin could increase the proliferation and survival ability of A2780 cells.3. N-cadherin could increase the cell-matrix adhesion ability, cell-cell adhesion ability, survival ability and mobility of A549 cells; E-cadherin could increase the cell-matrix adhesion ability and cell-cell adhesion ability, and decrease the proliferation, survival ability and the migration ability of A549 cells.
