OBJECTIVE:Bone Morphogenetic Protein2(BMP2) is one of members of Transforming Growth Factor β superfamily and one of growth factors. BMP2 is a kind of strong bone-inducing factor and playas crucial role in periodontal regeneration. There is enormaous potential value in clinical application. Tooth transplantation of allografts is a valuable clinical technology developed for long time,which has a great potential value in clinical applications. Though it's short-term achievement ratio is very high,but it's five years' long-term achievement ratio is not satisfactory and there really have many unsuccessful cases. We establish animal models to observe bmp-2's osteogenesis in tooth transplantation of frozen tooth allografts in mice,and analyze it's mechanism of action, contrasted with control groups, to evaluate if Bone Morphogenetic Protein2 can promote the concrescence of tooth transplantation of frozen tooth allografts in mice,and if it has statistical significance compared with control groups. To approach the process and the characteristic of tooth transplantation of frozen tooth allografts and to search a new method to promote the concrescence of tooth transplantation of frozen tooth allografts.METHOD:Sixty female 6-month-old SPF Wister rats were selected. The body weight was 300-350g.Twenty-four for preparing hypothermy frozen homogeneity variant teeth,while others were randomly divided into four groups to establish odontosteresis of mandibular first mola of rats: I group: Abv-bmp2/bioglass; II group: bioglass; IIIgrouop: Abv-bmp2; IV组: control group. After injected with 2% Ketamine Hydrochloride,they were local anesthetized with 2% Nocaine on the area of mandibular first mola of rats,then pull out the mandibular first mola of rats,take the variant tooth strained by 0.5g Abv-bmp2/bioglass into the alveoli- venter,sew up the gum exactly. In the second week, fourth week and eighth week,put the rats of everygroups to death, to observe the essay by X ray HE color and statistics analysis.RESULTS:1. In the second week,the main type of root resorption is inflamed resorption. There have no marked statistics difference among every groupd.2. In the fourth week, the main type of root resorption is vicarious resorption . There have marked statistics difference between Adv-BMP2 groups and no Adv-BMP2 groups. (P<0.0126)3. In the eighth week, the concrescence of periodontal membrane of the Adv-BMP2 groups are obviously higher than the groups without Adv-BMP2,and it has statistical significance;the vicarious resorption of the Adv-BMP2 groups are lower than the groups without Adv-BMP2, and it has statistical significance.(P<0.0126)4. In the eighth week,cementoid can be finded in the area of cemental,and resorption. alveolodental ligament can be finded union the neogenesis cementoid and neogenesis alveolar bone.CONCLUSION:1. Adv-BMP2 can be used to degrade the absorbing power of roots in the tooth transplantation of frozen tooth allografts in mice.2. In eighth week, the concrescence of periodontal membrane in the groups used Adv-BMP2 is higher than control groups without Adv-BMP2,and this han obviously statistical significance. That is to say Adv-BMP2 can improve the progress of the concrescence in the tooth transplantation of frozen tooth allografts in mice.3. Bioglass can not improve the progress of the concrescence in the tooth transplantation of frozen tooth allografts in mice,but maybe it is useful for neogenesis of the alveolar bone,it can be good to Adv-bmp2 for it's long effection.
