Objective: To advanced study the relationship between the gene expressions ofOrnithine decarboxylase and lung cancer from the level of RNA and protein. Itcan be used as the theoretical basis to the research of gene diagnose and genetherapy in the future.Method: RT-PCR technique is used to observe the density of ODC mRNA,next toobserve the difference of immunohistochemical. Western Blot technique is alsoused to observe the density of ODC mRNA. Final, MTT technique is used to observethe multiplication of different transfected cells.Results: The expression of ODC gene mRNA in lesion of lung cancer was significantly highter than that in contiguous normal mucosa (P<0.01), which is quietlyhighter in stage III than that in stage I and stage II (p=0.0095) .To someextent ,0DC recombinant vectors could inhibit the growth of A-549 lung cancercell lines .Conclusion: :ODC gene plays an important role in lung cancer and the up-regulatedexpression of ODC mRNA is related to the pathologic stages' of lung cancer. Tosome extent ,0DC recombinant vectors could inhibit the growth of A-549 lungcancer cell lines.
