Dental caries is one of the most common diseases. In all kinds of disease incidences, dental caries is in the front row. The newest data published by WHO in 2002-2003 indicated that although in developed countries dental caries incidence was keeping decrease in the last 30 years, it turned out to increase in some developing countries. Thus it is a very huge tusk for the prevention and cure of dental caries. CCL28( Mucosa Associated Chemokine) is a newly found CC family chemotatic factor, whose receptors are CCR10 and CCR3. CCL28 is mainly in salivary gland, prostate, lacteal gland, spleen and colon. A small quantity of CCL28 is also found in peripheral blood leucocyte and other mucosa tissues. Some researches had verified that CCL28 expressed highly in mice salivary gland. CCL28 is an effective chemoattractant for IgA antibody secrete cells (IgA ASC). It can express in salivary gland and colon. CCL28 attracts IgA ACS from all kinds of mucosa lymphatic organs and mucosa tissues exterior and interior colon. Besides, CCL28 has the character of antimicrobial activity and mucosa enriching, which composes the broad-spectrum protective layer together with other mucosa antimicrobial factors.In our early researches, we successfully expressed SBR fusion protein contained 6 histidines tag in JM109 (DE3), and we purified SBR fusion protein that could be used as recombinant antigen vaccine for protection and cure of dental caries. In this experiment, we adopted ELISA, RT-PCR and in-suit hybridization to detect the CCL28 expression in galndula angularis in mice which were injected with SBR fusion protein and irrigated with attenuated salmonella. Methods:1. The purified streptococcus mutans SBR fusion protein was injected in the glandula angularis area of BALB/C mouse as subunit vaccine. The specified antibodies were detected by ELISA. CCL28 mRNA expression in the glandula angularis was detected by RT-PCR. CCL28 expression in glandula angularis was detected by in situ hybridization.2. The experimental group mice were immunized by the transferred attenuated salmonella. The bacteria were irrigated into the mice's stomachs. CCL28 expression in glandula angularis was detected by in situ hybridization. Results:1. The ELISA results showed that the purified SBR could significantly induce the specified SBR-IgG (P<0.01) and SBR-IgA antibodies in saliva.2. The RT-PCR results indicated that the CCL28 mRNA expressions in the immunized mice were higher than the control groups.3. The in situ hybridization results showed that CCL28 expressions in the experimental groups were higher that control groups.Conclusions:1. The purified streptococcus mutans SBR subunit vaccine can induce significant antibody response via the inoculation in glandula angularis.2. The streptococcus mutans SBR subunit vaccine injected partly in glandula angularis can not only induce the antibody response, but also can enhance the expression of CCL28.3. CCL28 expression in glandula angularis increased when the attenuated salmonella was irrigated in the mice's stomachs.
