Ionizing radiation has been used in many fields with modern tech development, especially in medical field. It has become one of the important treatments for malignant tumor. However, it also produced some secondary effects, as follows leucocyte decreasing, immune function depressing etc. It always induce that some patients have to give up being treated. Consequently, it has become urgently subject how to prevent or to decrease the injury of normal tissue, at the same time ensuring the effect of tumor therapy. Most of radioprotectors used today is chemosynthesis drugs, it has good effects,but its application is limited because of adverse reaction. The natural plant posses distinct anti-radiation effects and low toxicities,becoming the hot spot of radiation protection medicine.As a traditional Chinese medicine, Fructus Shisandrae has been used for a long period, having a wide pharmacological function and showing very good anti-oxidation and anti-radical effect. It hasn't been reported about protective effect of Fructus Shisandrae extraction on the radiation damage. We studied protective effect of Fructus Shisandrae extraction on radiation damage of mice. The purpose of this experiment was to observe the protective effect of Fructus Shisandrae extraction on radiation damage of mice, to produce economicaland effective medicine and provide the experiment gist.The administration method of Male mice was intragastric administration in this study. Mice were dived into three groups: scilicet, negative control group, positive control group and protection group. The negative and positive control group were given isotonic Na chloride, and protection group was given extractive of Fructus Shisandrae ( 100mg/kg, 200mg/kg and 400mg/kg),once a day in seven days pre-irradiation. Mice in positive control and protection group were generally exposed to x-ray after the last administration. All indexes were measured in the third day after irradiation. Indexes include the spleen and thoracic gland index, the number of WBC, the content of MDA, the SOD activity in live and micronuclear rates of PCE in bone marrow, in order to observe the safety action of Fructus Shisandrae extraction for radiation damage in mice, and conclude the following results.1.Effect of Fructus Shisandrae extraction on organ index of mice exposing to x-raysThe spleen and thoracic gland index of the mice decreased obviously post- x-ray irradiation at a dose of 2.0Gy(p<0.01). But organ index elevated obviously after mice were prophylacticly given Fructus Shisandrae extraction at dosages of 200mg/kg (or)and 400mg/kg(p<0.05).And the protection degree steped up as the extraction dose climbed on. When the mice exposing to a dosage of x-rays at 1.0Gy, 2.0Gy and 3.0Gy respectively, the spleen and thoracic gland index descended accordingly. It's significantly different from negative control group. The spleen and thoracic gland index number wereraised significantly after the mice were given Fructus Shisandrae extraction at dosage of 400mg/kg .2.Effect of Fructus Shisandrae extraction on WBC of mice exposing to x-raysAfter exposing to X-rays at a dose of 2.0Gy, the number of WBC was decreased obviously comparing to negative control group(p<0.01). But the cell number was higher after mice were Prophylacticly given Fructus Shisandrae extraction at dosages of 200mg/kg (or)and 400mg/kg(p<0.05).When the mice exposing to a dosage of X-rays at 1.0Gy ,2.0Gy and 3.0Gy respectively, the number of WBC descended significantly. The cell number could be raised significantly after the mice were given Fructus Shisandrae extraction at dosage of 400mg/kg .3.Effect of Fructus Shisandrae extraction on liver anti-oxidize ability of mice exposing to x-raysThe mice exposing to 2.0 Gy x-ray, the SOD activity descended obviously in hepatic tissue, and the MDA content increased obviously, there were significant difference between positive and negative control group (p<0.01). The SOD activity of the 400 mg/kg drug prophylaxis group were obviously increased than positive control group (p<0.05). The MDA content of the 200mg/kg and the 400mg/kg drug prophylaxis group were obviously decreased than positive control group (p<0.05). After mice exposing to the 1.0 Gy, 2.0 Gy and 3.0 Gy x-rays, the SOD activity descended obviously in hepatic tissue, the MDA content increased obviously, there were significant differencebetween positive and negative control group (p<0.01). And along with the increment of X-ray dosage, the SOD activity descended obviously, the MDA content gone up obviously too. SOD activity raised and MDA content descended after drug prophylaxis of 400mg/kg, comparing with the positive control group and there were statisticly differentce (p<0.05).4.Effect of Fructus Shisandrae extraction on PCE marrow cell micronucleus of mice exposing to x-raysThe mice exposing to 2.0 Gy x-ray, of bone marrow increased obviously comparing with negative control group (p<0.01). Micronuclei of the 200mg/kg and the 400mg/kg drug prophylaxis group decreased obviously comparing with positive control group ( p<0.05 or p<0.01). After mice exposing to the 1.0Gy, 2.0Gy and 3.0Gy x-ray, micronuclei increased obviously comparing with the negative control group(p<0.01),And along with the increment of X-ray dosage, micronuclei increased. Micronucleus of drug prophylaxis group increased obviously comparing with positive control group (P<0.05)The results of above experiment showed that Fructus Shisandrae have a certain protective function to the mice exposing to x-ray, it can increase the number of Peripheral blood leukocyte of the mice exposing to x-ray and can obviously raise the index of spleen and thymus gland; and can suppress the generation micronucleus. It also can raise the SOD activity and reduce the MDA content in liver of mice exposing to x-ray.Maybe the mechanism is related with the following effects of the Fructus Shisandrae: Fructus Shisandrae extraction can improve the enzymatic activity of antioxygen system, thereby it could reduce the quantity of free radical which isharmful to the rhagiocrine cells, then it could reduce the damage of X-rays, raise the spleen and thoracic gland index number as well. Also it can cut down the damage of X-rays to hematopoietic system, and promote the recovery of peripheral blood hemogram, it can decrease the micronuclear rates in PCE induced by X-rays . Whether there are other protective mechanisms is needed researched.
