Along with the development of the theories and the methods of cardiopulmonary cerebral resuscitation (CPCR), the level of CPCR have being elevated obviously, but some estimates indicate that the survival rate after cardiac arrest is still lower than 10%. Epinephrine has been administed as a drug essatial for CPCR. Especially, high dose epinephine may enhance the success rate of resuscitation of sudden cardiac arrest. However, many investigations were demonstrated that high dose epinephrine may result in more serious sequela and reduce the survival rate after cardiac arrest. Recently ,more and more evidences have indicated that excitatory amino acids transports (EAAT) play very important roles in the process of neurotixic effect induced by glutamate.But there are rarely report on the changes of EAAT of brain tissue of cardiac arreast animal model.Therefore we carry out the experiment to observe expression of EAAT-2mRNA in the brain of cardiac arreast rats. Meanwhile we assume that when we salve sudden cardiac arrest, we can add another drug to improve ischemic state of brain, which caused by large amount of epinephrine and protect cerebral cell. So we can not only not influence the effect of cardiac-pulmonary cerebral resuscitation, but also can alleviate the durg damage to cerebral tissue. Therefore we also observe the influence of intervention effect of Guhong injection to expression of EAAT-2 and change of brain histomorphology in the experiment. The objective is to enhance the achievement ratio of CPCR.Methods: Forty Wistar rats (body weight 300+20g) were divided into four groups at random: Sham group rats were not caused asphyxial and endotracheal tubes were clamped to cause asphyxial cardiac arrest with the rest rats. Control group rats were treated with epinephrine (0.2mg/kg) i.v. Epinephrine were given again after an interval of 5 min if necessary. Administration group rats were injected guhong injection (1.8ml/kg) deliquationed to 1ml with 5% GS 3 days before experiment,other operation was the same to control group. Successfully resuscitated rats accepted 2-hour-period of intersive care. Histological feature of cerebral tissue were examined under light microscope and electron microscope. Polymerase chain reaction was applied to observed changes of mRNA expression of EAAT-2 in hippocampus.Results: 1.Under light and electron microscope, we find cerebral tissue morphological of sham group rats were approximately normal, while ischemic and hypoxia damage was obviously lightened in administration group compared to in control group. 2. The mRNA expression of EAAT-2 in hippocampus of control group rat was weaker than sham group ones,while it of administration group rat was enhanced compared to sham group. Conclusion: 1. Expession of EAAT-2mRNA degrades in hippocampus of cardiac arreast rats and EAAT-2 is involved in the process of brain tissue damage after cardiac arrest. 2. Guhong injection can enhance the EAAT-2 mRNA expessin in hippocampus tissue of cardiopulmonary cerebral resuscitation rats. 3. Guhong injection could evidently improve hypoxia and ischemic changes of heart and nerve cell.Subsequently, It is beneficial for cardio-cerebral resuscitation.
