Objective: Analysis expression of SRD5A2 gene and SKY gene in patients with hypospadias to identify possible molecular mechanism underlined hypospadias and any relation2 ship of the expression of SRD5A2 gene and SKY gene to hypospadias.Methods: Thirty-six blood samples from the patients with hypospadias were obtained from December. 2004 to April 2005 and was classified anterior, middle, posterior. DNA was extracted from blood leukocytes. Exons 4 of the SRD5A2 gene and the SRY gene were amplified by PCR. Use it synthesis the probe to hybrid with RNA of blood samples . Imaging of the results.Results: In the tape of SKY gene, all of tapes contrast with control group and there is no significant difference in the width , brightness, location. In the tape of SRD5A2 gene, the brightness of the tape of posterior is pale-green and the width is narrow. There is no significant difference in the width , brightness, location of the other tapes.Conclusion: There is no significant difference of the expression of SRY gene in patients with hypospadias. The expression of SRD5A2 gene reduces in posterior paitien .So the expression of SRD5A2 gene affects the differentiation of the external genitalia and may play a role in the etiology of hypospadias. Since no changes of expression of SRY gene was found , this study might suggest that the expression of SRY gene is not an important event in hypospadias.
