Diosmin's is 3,5, 7-Trihydroxy-4'-methoxyflavone--7--rutinoside, it is a kind of Flavonoids which was extracted and isolated from Chinese herb Galium verum L. by the author. This medicine can alleviate limbs edama with the functions of increasing vein tension , improving vein permeability , and improving microcirculation . There are many clinical treatment report about its affirmative curative effect and it has vast prospect of utilization.During this subject ,the author extracted diosmin which was regarded as the target compound from Chinese herb Galium verum L.with various chromatography method comprehensively including macroporous resin chromatography, polyamide chromatography, N-P and R-P silica gel chromatography and spherdex gel chromatography. Then it was judged by physics and chemical constant as well as spectrum analysis. An specific and sensitive HPLC method was established for the determination of diosmin in rat plasma and it can be used by the biopharmaceutical quantitative analysis of this medicine. The pharmacokinetics of diosmin was studied in rat, and the one compartmental model was determined after the data and C-T curve was analysized by the pharmacokinetics program 3P97.With the in vitro manner, the author used Equilibrium dialysis method to imitate the binding process between diosmin and plasma protein, and determined the concentration of diosmin in and out of the dialysis membrane, and determine the binding rate at last.Pharmacokinetics is a subject which descripts medicine's dynamic variation regularities in body with kinetics principle. Its fundamental analysis method has penetrated into biopharmacy, medicinal chemistry, clinical treatment and promote them at the same time. Diosmin was used to cure limds edama, geriatric acute hemorrhoid and other microcirculation illness comprehensively as the microcirculation regulator. This study evaluates the vivo process of diosmin which was regarded as lead compound primarily with pharmacokinetics principle and biopharmaceutical analysis method. The result provide reliable foundation for the diosmin's possible process of chemical structure transformation, dosage form choice and designing, quality evaluation of prescription, pharmacology, toxicology and clinical treatment. Diosmin ,as one of main activated ingredients in Galium verum L, its pharmacokinetics result lays foundation for new drug exploration of diosmin and Galium verum L's pharmacokinetics of single herb and multiherb on the basis of Galium verum L's definite activated substance .
