Objective: The present study was to explore the expression ofHSP10, HSP27 and HSP47 in polycystic ovary syndrome. We employedmouse early antral follicle culture approach to determine the Hsp10localization and expression levels during follicle development. The aimof this study was to demonstrate the effects of Hsp10, Hsp27 and Hsp47on the etiology and pathophysiology development of polycystic ovarysyndrome, and these should be taken into consideration in futurestrategies focusing on prevention and treatment of PCOS.Methods: We carried out realtime-PCR, Western blot analysis andimmunohistochemistry to define the experssion of HSP10, HSP27 andHSP47 by 3 PCOS ovaries and 3 normal ovaries. Mouse early antralfollicles (diameter: 220-250μm) isolated from ovaries of 20-day-oldfemale mice were cultured for 3 days in vitro, morphological growth andsteroid production during follicle culture were measured.Immunohistochemistry (IHC) was used to determine the Hsp10localization and expression levels during follicle development.Results:①HSP10 mRNA doesn't have significant differentialexpression between PCOS and normal ovaries(p>0.05); HSP10 protein has a high expression in normal ovaries(p<0.05), and it was highlyexpressed in all the cells of normal ovaries, but only highly expressed inthe theca cells of PCOS ovaries.②Both of HSP27 mRNA and proteinwere highly expressed in normal ovaries(p<0.05); HSP27 was highlyexpressed in the oocyte of normal ovaries.③Both of HSP47 mRNA andprotein were highly expressed in PCOS ovaries(p<0.05); HSP47 washighly expressed in the oocytes, granulosa cells and stroma cells of PCOSovaries.During 3 days culture, the follicles increased in diameter, thegranulosa cells proliferate and the antral becomes enlarged, and theproduction of E2 progressively increased. In the mouse early antralfollicles, Hsp10 staining was limited to oocyte specifically, Hsp10expression was significantly higher in all the cell types of follicle afterculture than before.Conclusion: All of HSP10, HSP27 and HSP47 have significantdifferential expression between PCOS and normal ovaries, HSP10defferently expressed during follicle development. These differentiallyexpressed proteins may contribute to the development of polycystic ovary,anovulation, hyperandrogenism, insulin resistance of PCOS. The mouseearly antral follicle culture approach can help to understand the role ofthese differentially expressed proteins in pathophysiological development of PCOS.
