Object:To determine the expression of cyclinD1 protein in patients with acute leukemia , investigate cyclinD1' s function and clinical significance in acute leukemia,and find a valuable new target in the diagnosis and treatment of acute leukemia .Methods: The expression of cyclinD1 mRNA was determined by realtime quantitative RT-PCR method in 51 patients with acute leukemia, among whom 21 were ALL and 30 were AML, newly diagnosed group of 23 cases, complete remission group of 16 cases, refractory or relapse group of 12 cases. Samples of benign hemopoietic patients were used as normal control (16cases) .The follow-up visit has been taken in 16 patient from newly diagnosed group ,amang whom 7 patients have been released after treatment while the others were not.Results:1.The expression of cyclinD1 in patients with Acute Leukemia was significantly higher than that in control group,and there was a significant difference (p<0. 05). 2.There was no significant difference in the expression of cyclinD1 (p>0.05)between ALL and AML. 3.The expression of cyclinD1 was diverse among the three experiement groups (p<0. 001), the highest level was in the refractory or relapse group while the lowest level was in the newly diagnosed group. 4. In the follow-up visit after treatment,the expression of cyclinD1 in patients who hasn' t released was higher than that in patients of remission, and there was a significant difference between the two groups (p<0.05). Conclusion: 1.The over-expression of cyclinD1 in Acute Leukemia has some effect on the origin of the disease. 2.The expression of cyclinD1 is not specific between ALL and AML. 3.CyclinD1 is correlated with progression of the disease in a certain degree, espicially with relapse. Dynamic observation with the level of cyclinD1 can be used to evaluate the treatment outcomes and act as a prognostic marker for Acute Leukemia. 4. Chemotherapy to the disease can reduce the expression of cyclinD1 in turn reduce the tumor load. This indicates that chemotherapentic agent can inhibit cyclinD1 gene amplification in some degree,thus cyclinD1 perhaps become a new target in the targeted therapy to Acute Leukemia.
