Studies On The Chemical Constituents Of The Extract From Ginkgo Biloba & The Phasic Studies On Maidenhair Tree's Endophytic Fungus

On one hand, in this article, we isolated and identified the chemical composition of the extract from Ginkgo biloba, on the other hand, we isolated and identified Fungus GB-1(3)'s secondary metabolite, and we found anticancer and anticoagulated blood activity in it.In the respect of chemical composition's researching, we adopted the method of silica gel column chromatography, polyamide column chromatography and Sephadex LH-20 column chromatography. We have identified 11 compounds by using chemical and spectroscopy methods.The 11 compounds are: Genkwanin, Isorhamnetin, Kaempferol, Quercetin, Bilobalide, Ginkgolide A B C, Rutin,β-sitosterol and Daucosterol.We analyzed and discussed the absolute configuration of ginkgolide by adopting both the method of circular dichroism spectra and the theory of Klyne's lactone sector rule. The result of our research shows that the absolute configuration of ginkgolide matches what was reported in references very well.In the respect of researching on maidenhair tree's endophytic fungus, we transferred the culture of the two kinds of fungus whose secondary metabolism product may contain the utility chemical composition in Ginkgol biloba. Finally, we've got 21 kinds of single strain.By using HPLC, we found that it is possible that strain C-1 ( 3 ) cultivated by Czapek medium contains bilobalide and ginkgolide B. But in scale-up, we didn't find those two composition. Possibly, it was because of 2 reasons: 1^st , the content was too low; 2^nd, bad reproducibility.We've got 2 compounds from the petroleum abstraction. They are Soyasapogenol B andβ-sitosterol. It is suspected that both of the two compounds are the composition of the medium.The chloroform abstraction of the C-1 (3) strain's secondary metabolite has anticancer activity. By using the method of high-flux screening, we found that the No.56 fraction which was isolated by silica gel column was a better inhibitor of Factor Xa. What is the composition of this abstraction calls for further research.The endophytic fungus GB-1 was identified as Fusarium oxysporum Schlecht by morphological observation and DNA sequence comparison.
