Objective: To study the expression and clinical significance of lymphatic vesselendothelial hyaluronan receptor LYVE-1 mRNA in endometrial carcinoma and itsperitumoral stroma.Methods: Expression of LYVE-1 mRNA were assessed by semi-quantitative reversetranscription-polymerase chain reaction (RT-PCR) in 52 cases of endometrialcarcinoma (T group), its corresponding peritumoral stroma (PT group), and 13 casesof normal endometrium (NE group).Results: (1) LYVE-1 mRNA were detected in all 3 groups. The expression level ofLYVE-1 in PT group was the highest(1.3020±0.1042)compared with the other 2groups. LYVE-1 mRNA level decreases in order in NE group and Tgroup(0.9889±0.1365 and 0.6928±0.1437 respectively).(2) In PT group, the LYVE-1 mRNA level was relevant to the surgico—pathologicalstage and the myometrial infiltration depth.The expression level in stageⅢ,Ⅳ((1.3686±0.1024) was higher than that instageⅡ,Ⅰ(1.2802±0.0951,1.2141±0.0872 resecptively). There was a significantdifference among the 3 groups(P<0.01). Besides, the expression level in myometrialinfiltration depth>1/2 group was higher than that in depth<1/2 group, and thedifference is significant(P<0.05), yet the expression of LYVE-1 mRNA wasirrelevant to the surgical-pathological grade and pathological subtype (P>0.05).(3) Lymph node positive tumors had a higher LYVE-1 mRNA level than nodenegative tumors. The difference between the two group is significant (P<0.05).Conclusion: (1) LYVE-1 mRNA were detected in all 3 groups. The expression levelof LYVE-1 in PT group was the highest, while it decreases in order in NE group andT group, indicating that lymphangiogenesis occures in the area of peritumoral stroma.(2) LYVE-1 mRNA level was relevant to the surgico—pathological stage and themyometrial infiltration depth, while it was irrelevant to the surgical-pathologicalgrade and pathological subtype. (3) The Lymph node positive group had a higher LYVE-1 mRNA level than nodenegative group, indicating that new formed lymph vessels may promote tumor cellmetastasis, and then increases the chance of distant metastasis.
