Experimental Study About Model Combining Disease With Syndrome Of Type 2 Diabetes Mellitus With Qi And Yin Deficiency

Objectives: To create animal models combining disease with syndromeof Type 2 Diabetes Mellitus (DM) with Qi and Yin deficiency and probeinto corresponding method and criterion about it.Methods: 60 Wister rats were randomly divided into 3 groups, whichare Normal control group, DM control group and group of Qi and Yindeficiency. The rats in the group of Qi and Yin deficiency wereperfused with Reticulatae Viride and Radix Aconity LaterailsPreparata. All the operation as mentioned above lasted for 4 weeks,and all rats in thegroup of Qi and Yin deficiency were verified withZhong Hui Tang Mai Kang Granule. The observation targets includedgeneral status of rats, blood sugar,blood-lipid, cortisol,insulin, free triiodothyronine (FT₃), Thromboxane B₂(TXB₂),6-keto-prostaglandinF_1α(6-keto-PGF_1α) and pathologicalexamination of pancreas, kidney, adrenal gland,thyroid gland.Results: 1. After the rats were perfused with Chinese herbs, therats in group of Qi and Yin deficiency showed symptoms oflistless,lassitude, enlarged tongue,less saliva in tongue; andthe common symptoms of DM, such as polydipsia, polyuria,emaciation,and so on. 2. The value of blood sugar,triglyceride (TG),totalcholesterol(TC),low density lipoprotein(LDL),cortisol, FT₃,Insulin,TXB₂ and 6-keto-PGF_1α in rats of Qi and Yin deficiency groupand DM control group was distinctively different from that of normalcontrol group;there were great differences between the value oftriglyceride (TG),total cholesterol(TC),low densitylipoprotein(LDL),FT₃,insulin and TXB₂ in rats of Qi and Yindeficiency group and that of DM control group. The results ofpathological examination about the pancreas,kidney,adrenal glandand thyroid gland in Qi and Yin deficiency group and DM control groupwere markedly different from that of normal control group and therewere no markedly differences between Qi and Yin deficiency group andDM control group. 3. After verification, the level of TG,TC,LDL,insulin and TXB₂ recovered to some extent and there were greatdifferences from that before verification.Conclusion It is feasible to create animal models combining diseasewith syndrome of Type 2 Diabetes Mellitus with Qi and Yin deficiency by the method of perfusing herbs with different properties andflavors on the basis of traditional type 2 DM animal models createdby the way of feeding high lipid and injecting streptozotocin.
