ObjectiveSince the concept of neural stem cells (NSCs) was declared by Reynolds and Richards in 1992, about its research has provided penetrating insights into the plasticity and regenerative potential of the brain over last decade. As soon as neural stem cells was discovered, many experts took it seriously and studied its biological behavior and characteristics which were described as follows. (1) self-renewal: Neural stem cells have a fabulous ability of multiplication and self-renewal, which can carry on karyokinesis ceaselessly under certain condition; (2) Other kind of cells can be produced through asymmetric mitosis; (3) Neural stem cells can differentiate to neuron, neuroglia and other nervous tissues. Because of these characteristics, it's feasible to take it into application for repairing nervous system, especially for central nervous system. Nevertheless, how to clarify the procedure of multiplication and differentiation stands in the breach to resolve.Stem cells have been isolated from embryonic as well as adult central nervous system (CNS).This study is discussing about:1,To master the method of culture of NSCs in vitro.2,To study the effect of BDNF on the growth and differentiation of neural stem cells from the young SD rats.MethodsNeural stem cells were dissociated from the brain of 1 week's Sprague-dawley rats. The expression of Nestin was examined at the 6th day in culture. Following 4days in culture, the immunocytochemical analysis was used to identify the expression of neuro-specific enlase (NSE) in culture cells, including the number of living-cells and the situation of neurits.Results1,Most of culture cells were Nestin-positive.2,Tow groups (bFGF,bFGF + BDNF) all promoted stem cells to grow and differentiate, bFGF +BDNF mostly promoted them to differentiation into NSE-positive cells, to compared with control group(P<0.01), to compared with bFGF group (p<0.05).3,In bFGF + BDNF group, not only promoted them to differentiation into NSE-positive cells (P<0.01), but also promoted the growth of neuritis (p<0.01).Conclusion1,bFGF can promoted the growth of neuron in the differentiation from neural stem cells into neurons.2,Under the effecting of bFGF, BDNF not only can promoted the growth of neuron more obviously,but also can promoted the growth of neuritis in numbers.
