Objective:This study was designed to explore the effect of Sinomenine on the prevension and treatment of experimental autoimmune encephalomyelitis(EAE)in rats.To understand the possible mechanism about effect of Sinomenine on EAE,and then to apporoach the Sinomenine's application in clinic.Methods:(1)Preparation for EAE model:females wistar rats were about 6 to 8 weeks old,immunized with guinea pig spinal cord homogenate(GP-SCH)plu complete Freund's adguvant(CFA)and pertussis vaccine(PV) to make up the model.(2)Experimental animals'grouping:Wistar rats were randomly divided into four groups: EAE group,Sinomenine group,prenison group and the blank group.There are 5 rats in the blank group, 15 rats in the other three groups.(3)The way to deal with the rats which had fall to ill:in the initial stage,the peak time and the paracmasia,selected the brain and spinal cord to make sample.undertaking HE and LFB staining,observed the pathology change under microscope.detect the expressin of IFN-γ, IL-12 by immunohistochemistry. (4)Experimental datas were analysed with the statistics software Spss11.5.Results:(1)The condition of the incident and scord in each group ratsThe rats in EAE group begin to fall ill after 11 days,the fiftieth day to the peak,After 21 days,begin to release.the clinic manifestations arerlosing weight, limbs palsy,even on the brink of death. The two intervention groups delayed 2 days,the condition is likely the EAE group,but the clinical situation is better than the EAE group and great disparity among them.(p<0.05)(2)Disease rates and pathology change in each groupsIn EAE group the rat is 93.3%,the Sin group is 40%,the prednisone group rat is 46.67%,the blank group have no incidence.The disease rate in the EAE group is higher than two intervention groups.among them have obviously disparity(P<0.05),have no difference between two intervention groups. inflammatory cell infiltrate is obviously in the fastigium time of three groups,but focus of infection in the EAE group is more than in the other two groups(P<0.05).In LFB myelin staining,we can see demyelination in the EAE sections.(3) The immunohistochemistry result in thress groupsIFN-γ,IL-12 have expression in three groups,the expression with one accord with the condition of EAE group,obviously disparity between the initial stage and catabasis(P<0.05),they are higher in the EAE group than in the intervention groups(P<0.05).Conclusion:(1) The expression of IFN-γ,IL-12 were increased with the development of EAE,which demonstrated that IFN-γ,IL-12 played pro-inflammation roles in EAE.(2) Sinomenine could effectively the invasion and pathogenetic condition of rats in EAE,which might be related to keeping the balance of Thl/Th2 cytokines,reduced the producte of IFN-γ,IL-12.
