Objective: To explore Cdk3 expression in hepatocellular carcinomacancer and study the role of Cdk3 in hepatocarcinogenesis, and provideexperimental and theoretical evidence for the early diagnosis ofhepatocellular carcinoma.Methods: 1. Cdk3 protein expression was detected in hepatic tissuesof 9 patients with primary hepatocellular carcinoma (HCC), 7 patientswith hepatic cirrhosis, and 10 patients with hepatitis usingimmunohistochemistry, Cdk3 mRNA was detected in hepatic tissues of 4patients with HCC, 6 patients with hepatic cirrhosis and 4 patients withhepatitis using hybridization in situ. 2. Cell colony was observed usingsoft-agar experimental to study Cdk3 expression for the effects ofmalignant transformation; 3.Co-localization of Cdk3 and c-Jun wasdetected by bicolor-immunofluorescence assay in JB6 to study the role ofCdk3 in hepatocarcinogenesis; 4. AP-1 activity regulated by Cdk3 wasdetected using luciferase reporter gene to research the role of Cdk3 inhepatocarcinogenesis.Result: 1. The expression of Cdk3 protein is in nuclear-kytoplasmtype, mainly in kytoplasm, it's expression increased significantly in HCC(P<0.05). The expression of Cdk3 mRNA is in nuclear-kytoplasm type, mainly in kytoplasm, it were expressed in hepatitis, hepatic cirrhosis andHCC, however, there was no significant difference (P>0.05). 2.Cdk3-JB6,Cdk3-DN-JB6 cell line were used to analysis the effect ofCdk3 over-expression for anchorage-independent growth, we found thatthe number of cell colony formstion was significantly higher in the Cdk3stability expression cell than that in Cdk3-DN cell (P<0.05) and no Cdk3transfected cells(P<0.05) in the stimulation of EGF in soft agar test; 3.Cdk3 combination with c-Jun play a role in JB6 nucleus and kytoplasm; 4.The AP-1 activity in Cdk3 stably-transfected cells was much greater thanthat in transfected with no Cdk3 or cells transfected Cdk3-DN (P<0.05).Conclusion: 1. Cdk3 protein expression increases in thecarcinogenesis process of HCC. This implys that Cdk3 plays an importantrole in hepatocarcinogenesis; 2. Cdk3 enhanced anchorage-independentgrowth in the stimulation of EGF; 3. Cdk3 co-localizes with c-Jun in thenucleus and kytoplasm; 4. Cdk3 promotes malignant transformation byup-regulating AP-1, these indicate that Cdk3 involves in hepatocellelarcarcinogenesis.
