[Objective] To observe the protection effects of resveratrol on SH-SY5Y cells oxidation damage induced by H2O2, and to explore its possible molecular mechanisms.[Methods] PD cell model was established by treating SH-SY5Y cells with 100μmol·L-1 of H2O2. SH-SY5Y cells were pre-treated with different concentration (0.5μmol·L-1, 5μmol·L-1, 50μmol·L-1, 100μmol·L-1, 125μmol·L-1, 200μmol·L-1) of resveratrol for 2h, then oxidation damage was induced by incubating the cells with 100μmol·L-1 of H2O2 for 8h. After the treatment, cell viability, MDA concentration and GSH-Px activity of each group were examined, mRNA expression level of different genes (Bcl-2, Bax, CAT, nNOS and COX-2) and protein level of COX-2 were also measured to understand underlying mechanism.[Results] The cell viability of SH-SY5Y cells decreased to 18.35% after being treated with 100μmol·L-1 of H2O2 for 8h, and the cell normal morphous were also disappeared, which suggesting that PD cell model was successfully established. Pre-treatment of resveratrol could effectively increase the cell viability of SH-SY5Y cells. In the resveratrol concentration range from 0.51μmol·L-1 to 100μmol·L<sup>-1, the cell viability increased from 14.36% to the peak of 74.36%. From 100μmol·L-1 to 200μmol·L-1, the cell viability gradually decreased. In the resveratrol concentration range from 0.5μmol·L-1 to 100μmol·L-1, the mRNA expression level of Bcl-2 increased with increase of resveratrol concentration, while the mRNA expression levels of Bax,CAT,nNOS,COX-2 and protein level of COX-2 decreased along with the increase of resveratrol concentration.[Conclusion] The PD cell model could be established by inducing SH-SY5Y oxidation damage with H2O2. Resveratrol could significantly protect SH-SY5Y cells from oxidation damage induced by H2O2. Rsveratrol may exert protection effects via modulating the expression level of several genes related to oxidative stress and apoptosis. Rsveratrol may have therapeutic potential in the treatment of neurodegeneration diseases such as Parkinson's disease.
