Objective:To investigate the expression and localization of growth differentiation factor 15 (GDF-15)in human colorectal cancer(eRe)and the relationship between the changes in GDF-15 protein & rnRNA levels and tumor progression of CRC.Method:The pathologic characters of CRC patients were determined by HE staining in all of the 31 pairs of specimens of the eRe tissues and their surrounding normal tissues(at least 5cm from cancers),while immunohistochemistry,RT-PCR and Western Blotting were performed to determine the GDF-15 mRNA & protein levels of specimens.Then the biological roles of GDF-15 in colorectal tumor progression and the relationship between changes in GDF-15 expression and characters of CRC were evaluated.Result:GDF-15 expression was noted in the cytoplasm of colorectal carcinoma ceils and some cellular mesenchymalis,and some in the surrounding matrix of cancer cells and even the glandular organs in normal tissues.Statistical results showed the positive rate of GDF-15 protein in colorectal carcinoma(74.19%)was significantly higher than that in normal tissue (9.68%,P<0.01)by immunohistochemistry and Western Blotting and GDF-15 mRNA level in CRC was higher than that in their surrounding normal tissues with RT-PCR(P<0.05). The levels of GDF-15 protein & mRNA in poorly differentiated CRC were significantly higher than those at in the well and moderately differentiated,but the level in the CRC with lymphaden metastasis positive was significantly higher than that in the CRC without lymphaden metastasis(P<0.05).However,there was no relation between the positive rate of GDF-15 protein & mRNA and locations & shapes of the cancers(P>0.05).Conclusions:In this study,we evaluated GDF-15 protein & mRNA levels in CRC were higher than those in their surrounding normal tissues,which was related to pathologic characters.GDF-15 protein & mRNA levels in CRC were related to lymphaden metastasis and direct correlated with Dukes' stage but negative correlated with the differentiation of primary tumor.Gene amplification,increase in mRNA transcription or decrease in mRNA degradation may lead to both increase in GDF-15 protein and mRNA.In a word,GDF-15 can play a key role in occurrence and development of colorectal carcinoma and may be an indicator of malignancy and development of tumor.GDF-15 would be the identification of accurate diagnostic markers and GDF-15 measurement would be a predictor of CRC cancer,which provided the foundation of theory and experiment on the new targets and strategies for the management of CRC.
