AR Expression In Mice With Hypospadias Induced By Dichlorvos

Object Hypospadias is one of the kinds of birth-defect which is routine monitored by international.Cause of the disease is multiple.But the dysfunctional androgen receptor(AR)are paying more attention recently.They're the co-passageway of many etiopathogenisis of hypospadias.This research intended to induce hypospadias modles with mice by phosphate pesticide—dichlorvos and test the sexual hormone and AR in skin of penis and testis of mice.To investigated the pathogenesy of hypospadias.Methods 1.To selected 60 mice(Kunming species),randomizated 6 groups:A,B,C,D,E and F,from the 12nd day of pregnancy,received gavage of dichlorvos for 6 days with a dosage of 0mg·kg^-1·d^-1,5mg·kg^-1·d^-1,10mg·kg^-1·d^-1,20mg·kg^-1·d^-1,30mg·kg^-1·d^-1and 40mg·kg^-1·d^-1.30 days after the delivery,we observed the morphologic change of the male whelps exterior genitalia and congenital hypospadias were edtablished.2.To selected 22 normal mice,22 non- hypospadias mice of modle group,and 22 hypospadias mice,seperatedly detected the testosterone(T)and estradiol(E₂)by radioimmunoassay.3.Random selected 10 male whelps from the 3 groups respectively,to take the content of AR by immunohistochemical method of skin of penis and testis.Result 1.different dose of dichlorvos has different influence of incidence of hypospadias in mice.The incidence rate of hypospadias were 9%(4/47),28%(10/36)and 36%(11/31)in group D,E and F seperately.In addition,the offspring of pregnant mice were decreased significantly in group E and F(P＜0.05).2.The contents of T and E₂ in the 3 groups had no distinction(P＞0.05). 3.The AR content in skin of penis in.mice with hypospadias was obviously lower than the other group(P＜0.05).The AR content of skin of penis in blank mice and non- hypospadias mice of modle group had no manifest difference(P＞0.05).4.In testis in mice with hypospadias there was not obviously difference of The AR content than the other group(P＞0.05).Conclusion Kunming species mice can be built up hypospadias modles successfully if gavage of dichtorvos for 6 days with a dosage of 30mg·kg^-1·d^-1and 40mg·kg^-1·d^-1.The mechanisms of phosphate pesticide—dichlorvos could induce mice hypospadias modles may be dichlorvos decrease the expression of AR of skin of penis,and this can influence the combination between androgen and AR.But the level of T and E₂ in mice with hypospadias did not changed significant.
