| Objective To investigate the effect and molecular mechanism of Mevalonic acid (Mev)on the expansion of Human Mesangial cell(HMC)and the mitogen-activated protein kinase(p38MAPK)signal pathway.Methods①HMC were randomly divided into control group and Mev group,and at the same time,the Mev group was divided into 1nmol/L,10nmol/L,100nmol/L,1μmol/L,10μmol/L,100μmol/L,1mmol/L.the degrees of HMC proliferation were examined at the 12h,24h,48h to the Mev group which is more proliferable to HMC.The degrees of HMC proliferation were measured by MTT.②HMC was randomly divided into control group,Mev group, Mev+SB203580 group and SB203580 group.Using the methods of ELISA,we detected the expression of TGF-βin the downstream of MAPK.the expression of Bax,Bcl-2 and P38 were measured by Western blot.Results①Mev was found to stimulate HMC proliferation in a dose-dependent and time-dependent manner.②Compared with the control group,the Mev group induced a sharp increase in the expression of TGF-β.There is a decline in the expression of TGF-βwhen the inhibitor SB203580 was put into the Mev group,but there shows little difference.③Compared with control group,there is an increase in the expression of p38 in Mev group but shows little difference.There is a sharp decline in the expression of p38 in the Mev plus SB203580 group compared with Mev group.④Compared with control group,Bax was declined and expressed in Mev group,and there is no significant expression can be seen between Mev plus SB203580 group and Mev group.⑤Compared with control group,the expression of Bcl-2 was increased and expressed in Mev group,and there is no significant difference can be seen in Mev plus SB203580 group and Mev group.Conclusion Mev was found to increase HMC proliferation in a dose-dependent and time-dependent manner..Mev can stimulates the expression of TGF-β,there is no obvious decline when SB203580 was put into the Mev group.there is no significant relation between p38MAPK pathway and the expression of Bax and Bcl-2.
