Objective:Matrix metalloproteinase-1(MMP-1)promoter gene is additional inserted Guanine G at transcribing beginning upstream site—1607bp site.The single nucleotide polymorphism(SNP)of MMP-1 gene can elevate MMP—1 transcriptional level.This is possible owing to the organism having 2G allele which binds with this section enhance MMP-1 transcription.In MMP-3 promoter region—1171bp site,there is a site of the single nucleotide polymorphism.The polymorphism at the site can change the promoter transcription activity,and accommodate MMP-3 expression level.The single nucleotide polymorphism(SNP)of Matrix metalloproteinase gene can accommodate MMPs expression such as MMP-1,MMP-3.Hypsi-expression of MMP-1,MMP-3 can increase degradation extracellular matrix(ECM),assigning the capability of tumor cell passing through basement membrane and surrounding tissue,which precipitates tumor vessel formation.And so facilitate malignant tumor invasion and metastasis.The objective of this experiment are(1)Investigation at gene level to judge whether MMP-1,MMP-3 SNP phenomenon in the promoter region has some correlation relationship with cancer of the cervix affectability,whether it can be considered as the risk factor of forecasting cancer of the cervix.(2)Approaching the dependability between SNP different genotype in MMP-1,MMP-3 promoter region and the clinical stage and the pathos-grading of invasion cancer of the cervix.Methods:Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) was used to detect MMP-1,MMP-3 genotype at promoter region SNP situs in 60 cancer of the cervix patients,100 healthy controls.To analyze the dependability between each genotype, cancer of the cervix susceptibility and ganglia lymphatic metastasis.We made use of SPSS13.0 software to carry out statistical analysis.X~2 test has been used to analysis the difference between MMP-1,MMP-3 two sets allele and the genotype distribution.The relative risk odds ratio(OR)and 95%confidence interval haved been calculated with X~2 test.We use it to analyze the dependability between each genotype and the cancer of the cervix susceptibility and the clinical stage,pathos-grading of cancer of the cervix. Results(1)The 1G/1G,1G/2G and 2G/2G genotypic frequency are 10.0%,23.3%,66.7%and 24%,40%,36%in the cancer of the cervix patients group and the healthy control group.The distribution between the two sets has significant difference(x~2=14.431,p=0.001);The 1G and 2G allele frequency are 21.7%,78.3%and 44%,56%,there is significant difference between the two sets(x~2=16.312,p=0.000).(2)The 5A/5A,5A/6A and 6A/6A genotypic frequency are 1.7 %,26.7%,71.7%and 3%,35%,62%in the cancer of the cervix patients group and the healthy control group.The distribution between the two sets does not have significant difference(x~2=1.618,p=0.445>0.05).The 5Aand 6A allele frequency are 15%,85%and 20.5%,79.5%,there is not significant difference between the two sets(x~2=1.509,p=0.219>0.05). Solitary 2G/2G or 2G/2G+1G/2G genotypes all can obviously increase the onset risk of cancer of the cervix,compared with 1G/1G genotype.The risk value are OR=4.444(95%CI=1.633~12.100),OR=2.842(95%CI=1.088~7.424).Solitary 5A/5A or 5A/6A+5A/5A genotypes can not increase the onset risk of cancer of the cervix,compared with 6A/6A genotype.The risk value are OR=0.481(95%CI=0.048~4.776),0.645(95%CI=0.323~1.288).Through R×C chi square test,we can neither presume MMP-1 genotype having some correlation with the pathos-grading nor with the clinical stage of cancer of the cervix(P=0.795,P=0.528).And we can neither presume MMP-3 genotype having some correlation with the pathos-grading nor with the clinical stage of cancer of the cervix.(P=0.963,P=0.448).Conclusion:(1)There is some correlation between 1G/2G polymorphism in -1607bp site of MMP-1 gene promoter region and cancer of the cervix susceptibility of Chinese north Han femme.2G allele can obviously increase the risk of troubled cancer of the cervix.(3)We can not presume the dependability between the single nucieotide polymorphism of MMP-1,MMP-3 and the parameters about p athos-grading and clinical stage invasion cancer of the cervix,which may hint that we still need more research to reveal the dependability between the single nucleotide polymorphism of MMP-1,MMP-3 and tumorous invasion and ganglia lymphatic metastasis.
