Bladder tumor is urogenital system frequent malignant tumor.At present,therapeutic tools of bladder tumor are the operation,chemotherapy,radiotherapy and immunization adjunctive therapy.After the operation,making chemistries drug pour into the bladde is important method to cure bladder tumor.However,after the chemotherapy,bladder tumor has 40%~80%recurrence rate.Multi-drug resistance(MDR)remains one of the major obstacles to successful chemotherapy.More than 90%of tumor patients died of MDR.Consequently,there is still an urgent need of a viable strategy to overcome MDR.Microwave is a high frequency electromagnetic waves.As high frequency and short wave length,microwave has several advantages as follows:(1)The warmup depth is big,Directionality is fine,Also can focus,get very narrow microwave beam.After human body organization part absorbs the microwave electromagnetism energy,what be come into being quantity of heat is not easy to give off to the outside.(3)It does not need any coupling agent when treating clinically.(4) microwave fever cures safety reliably,allocation is accurate,easy to handle.(5)equipment price is cheap.Be to reverse bladder tumour cell MDR ideal means as a result.In this study,we aimed to explore the possibility of using microwave for the purpose of MDR reversal,to select the optimized parameters of therapeutic microwave,and to investigate possible mechanism of microwave-induced MDR reversal.PART ONE Establishment and Characterization of Multi-drug Resistant Bladder Tumor T24 Cell linesObjective:To establish multi-drug resistant bladder tumor T24 cell lines and to assess their resistant characteristics.Methods:Bladder tumor T24 cell lines was exposed to adriamycin(ADM)in the culture medium for the establishment of drug resistant cell lines:concentrations of ADM was stepwise increased for long exposure.Morphologic studies were performed with optical microscopy.Drug sensitivities were determined by MTT assay,MDR1mRNA were detected by reverse transcriptase-polymerase chain reaction(RT-PCR).Results:Six months were taken to establish drug resistant cell linesT24/ADM.No obvious morphologic changes were observed between resistant and parental cell.But,drug resistance to ADM heighten.Resistance indices(RI)of T24/ADM to ADM was 15.79.RT-PCR showed that the expressions of MDR1 recreased remarkably.P<0.01 among all groups there are significant differences. Conclusion:Cell lines T24/ADM were multi-drug resistant.and it's drug resistance related to the overexpression of mdr-1 gene production P-gp.It is qualified to further research on multidrug reversing.PART TWOReversal Induced by microwave in Bladder Tumor T24/ADM CellsObjective:To observe Effect of microwave on Cell Viability and Reversal Induced by microwave in Bladder Tumor T24/ADM Cells.Methods:Use frequency 2450MHz,wavelength 12.24 cm microwaves irradiate the bladder tumour T24/ADM cell.The sensitivity of T24/ADM cells to adriamycin and reverse multiples were detected by MTT assay.Results:After microwave was used to reverse T24/ADM cells,a decreased IC50 from to 0.992ug/ml to 0.382ug/ml adriamycin on T24/ADM cells reversed as compared with nonreversed cells,drug-resistant index was 2.60.The part T24/ADM cells has been restored to chemotherapy medicine sensibility.P<0.01 among all groups there are significant differences.Conclusion:Microwave could partly reverse multi-drug resistance in the bladder tumor T24 cell cells.The mechanisms involved are as follows:①increase sensitivity of cells to chemotherapeutic agents;②facilitate the cellular uptake of drugs;increase intra-cellular medicine concentration.
