OBJECTIVE: Chronic renal failure (CRF) maintenance hemodialysis patients are associated with an increasing risk of ischaemic heart disease (IHD), but the mechanisms are not clear. Intercellular adhesion molecule-1 (ICAM-1) increased expression and Endothelial nitricoxide synthase (eNOs) decreased expression, the markers of endothelial cells dysfunction, have the relationship with ischemic heart disease (IHD). We investigated the effects about the expression of ICAM-1 and eNOs which maintenance hemodialysis patients' serum stimulated the human umbilical vein endothelial cells (HUVEC).METHODS: We adopt the outside development of HUVEC, and use the MTT method to investigate the influence of the maintenance hemodialysis patients' serum to HUVEC .The HUVEC are divided into random three sets :The 10 %fetal cattle serum (FCS) set (set A): use containing 10 %FCS 1640 liquids to incubate 1 hour,3 hours,6 hours and 12 hours respectively ;Normal persons' serum set(set B):Join to imply 10% normal persons' serum 1640 liquids to incubate 1 hour,3 hours,6 hours and 12 hours respectively ;Maintenance hemodialysis patients' serum set(set C): Join to imply 10% maintenance hemodialysis patients' serum 1640 liquids to develop for 1 hour,3 hours,6 hours and 12 hours respectively. Use the immunity cell chemistry method to investigate the expression of ICAM-1 and eNOs.RESULTS:1. The activity of HUVEC (the OD of MTT) is different stimulated by set A,set B and set C .The activity of HUVEC was decreased significantly in Maintenance hemodialysis patients' serum set (set C) after stimulating HUVEC 18h and 24h (P<0.05). And the inhibited rates are 24.92%, 43.68%. There are no differences of the activity of HUVEC between set A, set B and set C cultured 12h (Compared with control set P>0.05). 2. Compared with set A and normal patient set (set B) , the expression of ICAM-1 at lh was increased significantly in patients' serum set (set C) (P<0.05). The expression of ICAM-1 increased with the time at 3h, 6h and 12h (P<0.01). There was no difference in the expression of ICAM-1 between set A and set B at the same stimulated time (P>0.05) The expression of ICAM-1 increased at different stimulated time (P<0.01) . There was no difference in the expression of ICAM-1 between set A and set B at different stimulated time (P>0.05).3. Compared with set A and set B, the expression of eNOs at 3h was decreased significantly in set C (P<0.05). The expression of eNOs decreased with the time at 6h and 12h (P<0.01). There are no differences in the expression of eNOs between set A and set B at the same stimulated time (P>0.05). The expression of eNOs decreased at different stimulated time (P<0.01). There was no difference in the expression of eNOs between set A and set B at different stimulated time (P>0.05).CONCLUSION:1. Maintenance hemodialysis patients' serum can promote the expression of HUVEC's ICAM-1, and the cause the Endothelium cell dysfunction. With the serum stimulating time prolonging the expression of ICAM-1 obviously increased.2. Maintenance hemodialysis patients' serum can inhibite the expression of HUVEC's eNOs. With the serum stimulating time prolonging the expression of eNOs obviously decreased.
