Objective: In this study, we aimed at blocking the expression of multidrug resistance gene-1 in K562/A cells by RNA interference (RNAi). Methods: Chemical synthesized DNA oligonucleotide encoding the small hairpin RNA of mdrl gene was recombined into pSilencer4.1-CMV. The expression vector obtained eukaryotic RNAi was then introduced into K562/A cells with LipofectamineTM2000, and positive colonies were screened with Hygromycin B; The expression of mdrl in K562/A cells before and after transfection was detected by Western Blotting, RT-PCR and MTT method. Results: The expression of mdrl in K562/A cells was inhibited by the above small hairpin RNA of it. Conclusions: The specific small hairpin RNA targeting mdrl gene could inhibit the expression of this gene. For multidrug resistance reversal and treatment of leukemia this study results has theoretical and practical significance.
