Objective: To observe the morphological changes of atorvastatin pretreatment combining with ischemic preconditioning after cerebral ischemia-reperfusion injury, and in order to investigate more effective brain protective agent and preventive means.Methods: 32 wistar rats were randomly divided into four groups. A group was Sham-operated group; B group was ischemia-reperfusion group; C group was ischemia-preconditioning group; D group was atorvastatin preconditioning group, and the reversible focal cerebral ischemia-reperfusion models were set up by occluding the middle cerebral artery. After all operations were finished, pathological samples were made and fixed with cerebra Then the brains were cut into 2 ram-thick coronal sections at the level of the optic chiasm, including neocortex (most of them are frontal lobe and parietal lobe), preoptic area, piriform area, striate body, cornu ammonis and cerebral ganglion. Meanwhile cut them into 3 um-thick sections after paraffin imbedding. Finally, to observe the morphologic changes of cortex of frontal lobe, cortex of parietal lobe and cornu ammonis fields CA1 by using HE stain.Results: Neurons including A group of cortex of frontal lobe, cortex of parietal lobe and cornu ammonis CA1 fields had pyknosis necrosis and kytoplasm acidophilic degeneration slightly. Neurons including B group of cortex of frontal lobe, cortex of parietal lobe and comu ammonis CA1 fields compared with A, were dropsied distinctly, pyknosis necrosis and kytoplasm acidophilic degeneration, C and D groups were significant differences (P<0.01), the neuropil lose not clearly. C and D groups' pathological changes were nearly same (P>0.05), Neurons including cortex of frontal lobe, cortex of parietal lobe and cornu ammonis CA1 fields were dropsy lightly, pyknosis necrosis and kytoplasm acidophilic degeneration excessively, the neuropil lose not clearly. C and D groups' morphological changes compared with B groups', their damnifications were lighter than B group (P<0.01).Conclusion: Both Cerebral ischemia-preconditioning and atorvastatin pretreatment have protective effect after cerebral ischemia-reperfusion injury in rats. Neurons' sensibility that cornu ammonis CA1 fields is higher than cortex of frontal lobe and cortex of parietal lobe. The pyknosis necrosis changes of cortex of frontal lobe, cortex of parietal lobe and cornu ammonis fields CA1 neurons are upward of kytoplasm acidophilic degeneration with cerebral ischemia.
