Object: To overview neuritin expression profile in human cancer and corresponding normal tissues, and to observe the effects on cells apoptosis and senescence.Methods: (1) Immunohistochemistry(IHC) was used to screen neuritin expression in lung cancer, esophagus cancer, endometrial cancer, cervix carcinoma and corresponding normal tissues (5 samples in each tissue). And then baced on the detection of different expression of neuritin in cancer/normal tissue screened above, samples were added to 30 cases to confirm the results. The expression level of neuritin was detect in fresh tissues using RT-PCR. (2)β-galactosidase assay was used to detect the senescence of the BMSCs which expressed neuritin protein stably. And the flow cytometer was used to detect the apoptosis of BMSCs after being stained with Annexin V-PI.Results: (1) IHC screening results showed neuritin expression was higher in lung cancer, esophagus cancer, endometrial cancer, cervix carcinoma than that in corresponding normal tissues, especially in lung cancer(P<0.01). RT-PCR results showed neuritin expressed in lung cancer, but we did not detect neuritin expression in nomal lung. (2) Comparing with negative control BMSCs-3.1 (47.67±3.05%),β-galactosidase positive cells in BMSCs-neu were decreased (19.33±1.53%). And there was significantly difference between them (t=12.14, p<0.05). (3) Comparing with BMSCs-3.1(38.10±1.28%), the apoptosis ratio in BMSCs-neu was decreased (25.11±1.17%), And there was significantly difference between them (t=9.22, p<0.05).Conclusion: (1)The expression of neuritin was different in some tumor tissues and corresponding normal tissues. (2) neuritin overexpression delayed the senescence of BMSCs cells. (3) neuritin overexpression inhibited the apoptosis of BMSCs cells.In a word, these results indicated that neuritin play a critical role in cell apoptosis. It maybe involved in tumorigenesis and cancer development as a apoptosis inbhibitor.
