Objective To observe islets outcome, anti-apoptosis effect and islets activity ofUlinastatin and ATP in rat islets isolation, islets transplantation and effects after isletstransplantation under the kidney capsula.Methods The rats were devided into 3 groups according whether with the treatmentof Ulinastatin,a pancreatic trypsin inhibitor, and ATP during the islets isolation process.The purified islets by Ficoll 400 uncontinuius-density gradient were compared inislets outcome, apoptosis status of islet cells (by TUNEL and FCM), activity of isletsand effects after islets transplantation between the groups.Results The outcome and anmouts of insulin release under the glucose stimulating ofisolated and purified islets in Ulinastatin group were superior to those in the controlgroup(P<0.01). The number of apoptosis islet cells in Ulinastatin group were fewerthan those in control group(P<0.01). The anmouts of insulin release under the glucosestimulating of isolated and purified islets in ATP group were superior to those in thecontrol group(P<0.01). The number of apoptosis islet cells in ATP group andUlinastatin group were fewer than those in control group(P<0.01).After isletstransplantation the ATP group had better control effect in glycemia than in thecontrol group.Conclusions With the treatment of pancreatic trypsin inhibitor Ulinastatin during theislets isolation and purification may increase the outcome of islets and protect theislets from trypsin digestion and prevent apoptosis of islet cells. With the treatment ofATP during the islets isolation and purification may prevent apoptosis of islet cellsand maintain the activity of islets and had better control effectin glycemia after isletstransplantation under the kidney capsula.
