| Objective: Our experiment is to study the effects of Shan He Jiang Zhi Medicament(SHJZM) on weight-reduction and fat-decrese in adiposity rats, to observe the changes of leptin, adiponectinand prove SHJZM can affect adiposity rats through modulate leptin, adiponectin excretion in adipocyte.Methods: There are 70 male SD rats, 55 rats were given a high-fat diet to induce obesity, the other 15 rats were given a standard diet as control respectively for 8 weeks. After the models were prepared success fully, they were randomly divided into four groups:low dosage, middle dosage, high dosage, obesity control. After 8 weeks interference with SHJZM, the weight and the naso-anal length of each rat were recorded and Lee's index were calculated. The TG, TC, LDL, HDL of each rat were carefully deter mined. The ELISA was used to determined the level of serum leptin in rats, the techniques of RT-PCR were adopted to test the gene expression of adiponectin in adipose tissues of rats.Results:1 Compared to the high-fat diet(HFD)group, the body weights, the Lee's indexes, and the weight of fat tissues of SHJZM groups significantly decreased(P<0.05).2 Compared with the HFD group, serum TG, TC, LDL of SHJZM groups were significantly decreased(P<0.05). HDL of the HFD group was no significantly difference with those of SHJZM groups(P>0.05).3 Compared with HFD group, leptin of SHJZM groups significantly decreased(P<0.05).4 Compared with HFD group, adiponectin mRNA expression of SHJZM group significantly increased(P<0.05).Conclusions:1 SHJZM could significantly lower body weight and the serum TG, TC, LDL of obese rats.2 The effects of SHJZM in reducing blood leptin may act as one of the actors in lowering body weight of the obese rats. 3 The effects of SHJZM in raising the adiponectin mRNA expression of fat tissues may act as one of the actors that result in lowering body weight in obese rats.
