Objective: To study anti-tumor and affecting immune cells activities of flavonoid compounds from Chrysosplenium nudicaule Bunge.Methods: (1)MTT assay was used to examined the effect of DTFG on the proliferation of human leukemia cells K562 and stomach cancer cells SGC-7901, and the effect of TTF on the proliferation of human multidrug resistant leukemia cells K562/ADM and stomach cancer cells SGC-7901. (2)Found out the apoptotic evidences of stomach cancer cells SGC-7901 induced by TTF by means of fluorescence microscope, electron microscopy, flow cytometry and Agarose gel electrophoresis. (3)Build guinea pigmodel of type I hypersensitivity and isolated lymphocyte and EOS. (4)MTT assaydetected the effects of DTFG and TTF on activities of lymphocyte and EOS isolated from guinea pig.Results:1.The growth of leukemia cells K562 and stomach cancer cells SGC-7901 was not inhibited by different dose of DTFG (P>0.05).2. The growth of multidrug resistant leukemia cells K562/ADM was inhibited by different dose of TTF after treated for 72h, the highest inhibitory rate wass 52.82%. The IC50 respectively was 300.31mg/L,170.28mg/L,65.69mg/L,19.81mg/L after the drug acted on multidrug resistant leukemia cells K562/ADM for 241k 481k 721k 96h.3. The growth of stomach cancer cells SGC-7901 was inhibited by different dose of TTF after treated for 48h, the highest inhibitory rate was 80.36%. OD of drug groups was different from control group (P<0.01). To compare a drug group with another, the difference had statistical significance (P<0.05). The IC50 respectively was 142.17mg/L, 29.23mg/L,8.33mg/L,9.75mg/L after the drug acted on stomach cancer cells SGC-7901for 24h,48h,72h,96h.4.The stomach cancer cells SGC-7901 that treated with TTF was observed morphologiccharacteristics of apoptotic cell by fluorescence microscope and electron microscopy. Thestomach cancer cells SGC-7901 that treated with TTF for 48h, flow cytometry analysisshowed that the proportion of G0/G1 phase was higher than that untreated (P<0.01) andapoptosis peak was formed, the highest apoptosis rate was 40.4%. DNA Agarose gelelectrophoresis showed a mark of "ladder" band.5.It was lower that the OD of lymphocyte in guinea pig blood after treated with DTFG for72h and lymphocyte in guinea pig peritoneal fluid after treated with DTFG for 48h thanuntreated (P<0.01).6.TTF had no effects to the activities of lymphocyte in guinea pig blood (P>0.05). TTF atdose of 4mg/L could bring down the activities of lymphocyte in guinea pig peritonealfluid. It was higher that the OD of EOS in guinea pig blood and peritoneal fluid treatedwith TTF for 24h and 72h than control group (P<0.01).Conclusion:1.DTFG could not inhibit growth of leukemia cells K562 and stomach cancer cells SGC-7901.2.In vitro, DTFG could inhibit the activities of guinea pig lymphocyte. 3.TTF had a weak inhibition to multidrug resistant leukemia cells K562/ADM and strong inhibition to stomach cancer cells SGC-7901. 4.TTF could induce the apoptosis of stomach cells SGC-7901. 5. In vitro, TTF could increase activities of EOS.
