Perimenopause is an important period for women. It has well known that postmenopause women show the lower level of estrogen, thus leading to systematic metabolic disorder, especially for serum lipid disorder. Most women become obesity or overweight for this reason, as well as the incidence of cardiovascular diseases increase, such as diabetes, cardiopathy, hypertension and so on. Hormone replacement therapy (HRT) can treat these disease and prevent obesity, but increase the incidences of ovarian cancer and endometrial cancer if it was used for long time. So it has become a focus of medicine scientific researcher to find a natural product, which had few side effects. It is reported that Schizandre Chinensis Baill (SCB) has estrogen activity, but few reports are available about its effects on obesity and cardiovascular diseases in postmenopause women.To simulate metabolism of female in perimenopause and postmenopause, ovariectomized (OVX) rat model was made in this experiment. The effects of SCB' water extract on body weight, serum lipid, leptin in ovariectomized (OVX) rat were investigated, and its mechanisms were explored. Female Sprague-Dawley rat aged three months were randomly divided into sham-operated(SHAM)group, ovariectomized(OVX)group, OVX+ SCB group according to body weight about 220±2 gramme. During the experiment, SHAM group and OVX group drank water , OVX+ SCB group drank water extract from SCB,SCB concentration was increased gradually from1% to 12%. Food and water were measured every day, all groups are free to drink and eat. Body weight were recorded every other 10 days. On the 60 day after surgery, all rats were fasted for 12 hours and then were killed. Serum levels of lipids, Hepatic function, blood glucose, Insulin, Estrogen, Leptin, Tumor Necrosis Factor-α(TNF-α), Interleukin-6(IL-6), Maleic Dialdehyde (MDA) and Total Anti-Oxidation Competence(T-AOC) were assayed. All data were statistically analyzed by independent sample t test ((?)±s) with the computer statistical pakage SPSS13.0 .difference with P<0.05 were considered significant.Our experimental results were showed as follows: compared with SHAM group, rat in OVX group displayed increased body weight, Lee's index, food intake, Leptin, TNF-α, MDA, and serum level of lipids .While the water extract of SCB were given to the OVX+ SCB group, all of the results above have been impoved obviously. Our results indicate that SCB has estrogenic activity, which can inhibit food intake and body weight gain; improve the disorder of serum lipid metabolism, such as decrease serum levels of TC,TG,and HDL-C; regulate the adipocyte cytokine, such as decrease serum levels of leptin and TNF-α; enhance Anti-Oxidation Competence, such as decrease serum levels of MDA in ovariectomized rats.
